Embodiment Construction

[0033] In order to make the purpose, technical solution and advantages of the present invention clearer, the present invention will be further described in detail below in conjunction with the accompanying drawings and embodiments. It should be understood that the specific embodiments described here are only used to explain the present invention, not to limit the present invention.

[0034] The near-infrared fluorescence resonance energy transfer (FRET) sensor provided by the present invention uses near-infrared quantum dots (NIR-QDs) modified by nucleic acid aptamers (Aptamer) as energy donors, and oxidized carbon nanoparticles (OCNPs) as label-free energy The acceptor realizes the self-assembly of the donor acceptor through the π-π interaction. The sensor is based on FRET technology, and near-infrared light is used as the detection window, which overcomes the interference of background fluorescence and scattered light when visible light is used as the detection window, so th...

Abstract

The invention provides a construction method for a near infrared fluorescence energy transfer based biosensor. The method includes: subjecting a nucleic acid aptamer to covalent coupling to a fluorescent donor surface, mixing the nucleic acid aptamer coupled fluorescence donor with a label-free fluorescent receptor till fluorescence quenching of the fluorescence donor, thus obtaining the near infrared fluorescence energy transfer based biosensor. As the sensor detection window is located in a near infrared region, the background fluorescence, the background fluorescence, scattered light, light absorption and other optical interference can be well overcome, and the detection sensitivity is improved. The biosensor can be directly applied to complex biological matrix analysis.

Description

technical field [0001] The invention relates to the technical field of spectrum application, in particular to a construction method of a biosensor based on near-infrared fluorescence energy transfer. Background technique [0002] Fluorescence resonance energy transfer (FRET) is a highly sensitive, high spatial resolution, non-destructive homogeneous detection technology that combines fluorescence spectroscopy with resonance energy transfer technology. FRET means that the fluorescence donor and acceptor are in the range of 1-10nm. When the emission spectrum of the donor overlaps with the absorption spectrum of the acceptor, due to the dipole-dipole interaction, the excited state energy of the donor is in the form of non-radiation delivered to the recipient. This technology has been used in the field of analytical chemistry for more than 40 years, and it has greatly promoted the development of biomedical analysis while continuously improving itself. [0003] However, as a sp...
