Problems solved by technology

Although siRNA can efficiently and specifically inhibit gene expression in mammalian cells, the transfection efficiency of artificially synthesized RNA with general eukaryotic plasmid vectors is low, and it is difficult to become a gene therapy drug, and it is also difficult to achieve the purpose of gene interference

Abstract

The invention discloses a targeted rat phospholipase Cgamma2 interfering recombinant adenovirus and a construction method thereof. The targeted rat phospholipase Cgamma2 interfering recombinant adenovirus is obtained by inserting oligonucleotide shRNA (short hairpin ribonucleic acid) of targeted rat phospholipase Cgamma2 into an adenovirus genome-containing framework plasmid pHBAd-BHG, wherein the nucleotide sequence of the oligonucleotide shRNA is as shown in SEQ ID NO.1. Aiming at a siRNA (small interfering ribonucleic acid) nucleotide sequence of the rat phospholipase Cgamma2 gene, the invention designs and synthesizes the corresponding shRNA oligonucleotide sequence based on an adenovirus shuttle vector; the shRNA of the targeted target gene is continuously produced in a target cell by means of a U6 promoter of RNA (ribonucleic acid)-dependent polymerase III carried by the adenovirus vector itself; and the shRNA can be processed into the siRNA by Dicerase in the cell, thereby playing a gene interfering role.

Description

technical field [0001] The invention belongs to the field of biotechnology construction, and in particular relates to a recombinant adenovirus targeting rat phospholipase Cγ2 interference, a construction method and application. Background technique [0002] Acute liver failure is a group of clinical syndromes caused by severe liver damage caused by various factors, which in turn lead to serious dysfunction of the synthesis, detoxification, excretion and biotransformation of the liver itself. High (up to 80%), currently there is no specific treatment. The main pathological change is the massive death of liver cells. It has been found that one of the main forms of liver cell death in acute liver failure is apoptosis, and the occurrence of acute liver failure is closely related to liver cell apoptosis. Apoptosis, also known as programmed cell death, is an extremely complex process regulated by genes and plays an important role in the occurrence and development of liver failur...
