Embodiment 1

[0020] Example 1 Correlation between the expression of target microRNA (microRNA-676-3p, microRNA-181b-5p, microRNA-3591-3p) in plasma samples and the pathogenesis of AMS

[0021] 1. Material and Specimen Collection Description

[0022] Plasma samples of AMS patients were collected from 37 AMS patients who entered the plateau rapidly. Plasma samples of normal people were collected from 31 normal healthy people who entered the plateau rapidly. The diagnosis of AMS was confirmed by the International Common Diagnostic Criteria-Lake Lewis Score. AMS patients were not taking any medications before blood was drawn. A total of 2ml of blood was collected in EDTA-Na anticoagulant tubes for each sample.

[0023] 2. Sample processing and RNA extraction

[0024] Within 10 minutes after venous blood collection, centrifuge at 3000×g at 25°C for 10 minutes, use RNase-free and bacteria-free tips to take the upper plasma layer and store it in RNase-free and bacteria-free EP tubes at -80°C ...

Abstract

The invention relates to a microRNA marker for detecting acute mountain sickness, and application thereof to preparation of a kit for detecting acute mountain sickness. The content of microRNA-676-3p, microRNA-181b-5p and microRNA-3591-3p in the blood of a testee is detected by means of the quantitative PCR method and the like, and is compared with the normal microRNA level to diagnose acute mountain sickness. The acute mountain sickness molecular marker microRNA has high sensitivity and specificity, operation is convenient, safe and nondestructive, mass screening can be achieved easily, and the effect of combined use is better.

Description

technical field [0001] The invention relates to the technical field of medical molecular biology, and relates to the use of microRNA in blood plasma as a biomarker for the diagnosis of acute mountain sickness. Background technique [0002] Acute mountain sickness (acute mountain sickness, AMS), also known as acute mild altitude sickness, is a body reaction such as headache, loss of appetite, nausea, dizziness and fatigue caused by people who have not adapted to the natural environment of the plateau. It is the most common high-altitude idiopathic disease among people who rapidly enter the high altitude. According to existing studies, the incidence rate of AMS is 16-100% due to the age of the population, the altitude of entering the plateau, the speed and the diagnosis method of AMS (MacInnis M J et al. (2013) A prospective epidemiological study of acute mountain sickness in Nepalese pilgrimsascending to high altitude (4380m), "PLoS One", 8(10), e75644., McDevitt M et al. (2...
