Abstract

The invention discloses a preparation method of a mounting medium capable of simultaneously marking a cell nucleus. The preparation method comprises the following steps: 1) preparing 1 mol / L of TrizmaBase; 2) preparing 1 mol / L of NaH2PO4.2H2O; 3) preparing a Tris-phosphate buffer; 4) preparing glycerinum with the concentration of 50%; and 5) preparing the mounting medium. According to the prepared mounting medium capable of simultaneously marking the cell nucleus, the steps of dyeing and washing are omitted, the experiment flow is effectively simplified, the time cost is reduced, and the working efficiency is improved. The mounting medium can be used to mark and mount the cell nucleuses of samples like a frozen section, a paraffin section, a cell coverslip and a blood smear. The mountingmedium not only can be used to observe through a fluorescence microscope, but also can be used to observe through a laser scanning confocal microscope.
