Embodiment 1

[0018] Collect 50 cases of clinical fresh tumor specimens from the hospital, use IHC (immunohistochemical) method to observe the expression level of CDK5 in different colorectal cancer tumor tissues after fixing with 4% PFA, and use ImagePro Plus software to detect and analyze CDK5 in different tissues distribution in .

[0019] The results show:

[0020] (1) The expression of CDK5 (yellow brown area) in tumor tissue is much higher than that in paracancerous tissue, see figure 1 .

[0021] (2) The distribution area of CDK5 in tumor tissue is 16.5 times that of normal tissue, see figure 2 ;

[0022] The above results indicated that overexpression of CDK5 promoted the formation of colorectal cancer tumor tissue.

Embodiment 2

[0024] In order to verify the effect of CDK5 on the invasion ability of SW480 and DLD-1 cells, we verified it by Transwell chamber experiment.

[0025] After SW480 and DLD-1 cell lines (MOCK, sh-control, sh-CDK5 1#, sh-CDK5 5#) cells were continuously cultured under the same conditions for 48 hours, randomly selected fields of view were taken under the microscope and the number of cells passing through the filter membrane was counted .

[0026] result, see image 3 and 4 ,show:

[0027] The control group had more transgressed cells, while the sh-CDK5 1# group and sh-CDK5 5# group had fewer transgressed cells, and the experimental group was statistically significant compared with the control group.

[0028] The Transwell chamber experiment proved that cdk5 knockdown can significantly inhibit the invasion ability of tumor cells SW480 and DLD-1.

Embodiment 3

[0030] DLD-1 cells were treated with different concentrations of CDK5 inhibitor (Dinaciclib SCH727965), and then the expression levels of different cycle proteins in DLD-1 cells after cdk5 inhibition were detected by WB.

[0031] result, see Figure 5 , showed that: with the increase of drug concentration, the expression of P53 in cells increased significantly; the expression of P-PRb and P21 decreased significantly, but had little effect on the expression of Rb, which was consistent with the results of the knockdown group.

Abstract

The invention provides application of a CDK5 inhibitor to preparation of drugs used for promoting the expression of P53 in colorectal cancer cells. The invention also provides application of the CDK5inhibitor to preparation of drugs used for inhibiting the expression of P21 in colorectal cancer cells. The invention further provides application of the CDK5 inhibitor to preparation of drugs used for inhibiting RB phosphorylation in colorectal cancer cells. The invention further provides application of the CDK5 inhibitor to preparation of drugs used for treating colorectal cancer. The inventionfurther provides application of the CDK5 inhibitor to preparation of a kit used for detecting tumor markers. The invention discloses the regulation and control effect of CDK5 on the migration, invasion and transfer of colorectal cancer cells and the influence of CDK5 on a P53 signaling pathway, so the CDK5 inhibitor can be used as an effective target for treatment of the colorectal cancer and as anovel auxiliary diagnosis tumor marker; and thus, a novel method is provided for cancer diagnosis.

Description

technical field [0001] The invention belongs to the field of biomedicine, and particularly relates to the application of CDK5 inhibitors in the preparation of medicines for treating colorectal cancer. Background technique [0002] At present, the treatment of colorectal cancer mainly includes surgery, radiotherapy, chemotherapy, endocrine therapy, targeted therapy, and traditional Chinese medicine. However, since most of the tumors are found in the late stage, and postoperative recurrence is prone to occur, the curative effect of adjuvant therapy is not ideal, which affects the patient's survival and cure effect. However, at present, the effect of targeted therapy is still not good, and it is not effective for all patients, and the scope of application is narrow, and only patients with positive molecular target detection can apply targeted drug therapy. Contents of the invention [0003] Technical problem: In order to solve the defects of the prior art, the present invent...
