Embodiment

[0200] Solvents and reagents used for the syntheses were reagent grade from commercial suppliers and were used without further purification unless otherwise stated. Anhydrous CH 2 Cl 2 Grubbs, R.H.; Rosen, R.K.; Timmers, F.J. All humidity-sensitive reactions were performed under nitrogen or argon atmosphere using oven-dried glassware. Reactions passed in Kieselgel60F 254 TLC on the plate was monitored by UV, potassium permanganate and phosphomolybdic acid staining detection. Flash column chromatography was performed using silica gel (particle size 40-63 [mu]m). 1 H NMR and 13 C NMR spectra were recorded on Bruker AMX300MHz, Avance-500MHz and Avance-600MHz machines. Coupling constants are measured in Hertz (Hz) and spectra were acquired at 298k unless otherwise stated. Mass spectra were recorded on a Thermo Finnegan MAT 900XP, Micromass Quattro LC electrospray and VG70-SE mass spectrometer. Infrared spectra were recorded on a Shimadzu FTIR-8700 spectrometer.

Abstract

A lipid comprising a tri-chain cation of formula (la) having a cationic head group and three C12-24 hydrocarbyl groups for use in non-viral gene delivery systems, for example in the formation of lipopolyplex transfection vectors. Exceptionally good nucleic acid transfection is observed when nucleic acid and targeting peptides are formulated with the lipid of the invention (or lipid formulated witha co-lipid) into a LPD complex.

Description

field of invention [0001] The present invention relates to lipid derivatives suitable for use in the delivery of biologically active substances (eg, nucleic acids, proteins or small molecules) to cells. The invention also relates to complexes for use in the preparation of non-viral vectors for the delivery of biologically active substances to cells, said complexes comprising such lipids, such lipids in combination, for example, with peptides, and such complexes Use in eg prophylaxis, therapy and vaccination or in an in vitro laboratory setting. Background of the invention [0002] Gene delivery for therapeutic or other purposes is well known, especially for the treatment of diseases such as cystic fibrosis and certain cancers. The term refers to the delivery of a gene or part of a gene into a cell to correct some defect. In this specification, the term is also used to refer to any introduction of nucleic acid material into target cells, and includes both genetic vaccinatio...
