[0038] A method for evaluating ADSCs for improving hypertension in offspring rats, comprising the following steps:

[0039] (1) Cell culture: ADSCs were cultured in complete medium (basal medium + 10% serum + double antibody and glutamine); ADSCs were purchased from Saiye Biotechnology Company;

[0040] (2) Cell labeling: Dilute 1mg / ml SPIO with complete medium to 50μg / mL; at the same time, dilute polylysine to 1.5ug / ml, and then mix the above two solutions 1:...

Abstract

The invention relates to the technical field of biomedicine, in particular to an evaluation method of ADSCs for relieving filial generation rat hypertension and application. The method comprises the following steps of (1) cell culture: putting the ADSCs into a complete culture medium for culture; (2) cell marking: diluting 1mg / ml SPIO by a complete culture medium into 50 mug / mL; meanwhile, diluting polylysine into 1.5ug / ml; then mixing the two kinds of solutions according to a ratio of 1:1, wherein the final dilution concentration of the polylysine is 0.75 mug / mL; after the sufficient and uniform mixing, putting the materials into a cell culture box to be cultured; (3) pregnancy period modeling: building an LPS group, an LPS+ADSCs group and a control group (for the LPS group, injecting LPSinto the abdominal cavity of SD pregnant rats on the 8th day, 10-th day and 12-th day from the pregnancy; for the LPS+ADSCs group, injecting LPS into the abdominal cavity of SD pregnant rats on the 8th day, 10-th day and 12-th day from the pregnancy; meanwhile, injecting the ADSCs prepared in the step 2 into the caudal vein on the 8th day from the pregnancy; for the control group, injecting the equal volume normal saline into the abdominal cavity of SD pregnant rats on the 8th day, 10-th day and 12-th day from the pregnancy); 4, SPIO-ADSCs migration observation under MRI; (5) filial generation rat blood pressure change observation.

Description

technical field [0001] The invention relates to the technical field of biomedicine, and the specific field is an evaluation method and application of ADSCs for improving hypertension in offspring rats. Background technique [0002] Essential hypertension is a major risk factor for cardiovascular diseases and poses a serious threat to human health. Once hypertension is discovered, patients often need to take medicine for life, which not only reduces the quality of life of patients, but also brings serious social and economic burdens. Although extensive studies on its etiology and prevention have been carried out at home and abroad, its incidence rate is still increasing day by day, and the prevention effect for adult patients is not ideal, which strongly suggests that the current prevention and treatment ideas only for adult individuals with the disease need to be broadened. Indicates the presence of other important pathogenic factors. Therefore, it is increasingly importan...
