Embodiment

[0052] 10 kinds of polyphenols in fresh tobacco leaves (kaemferol, cyanidin, delphinidin chloride, rhamnazin, chlorogenic acid ), delphinidin 3-o-b-d-glucoside chloride, procyanidine B2, keracyanin chloride, rutin, scopoletin) As an example, the detection method provided in this application is described in detail as follows.

[0053] (1) Extraction of fresh tobacco leaves after pretreatment

[0054] First, take 50 mg of fresh tobacco leaves infected by TMV, freeze them with liquid nitrogen, freeze-dry them, and crush them with a pulverizer (200 mesh) for later use;

[0055] Secondly, put the pulverized tobacco leaf sample in a centrifuge tube, add 1.5 mL of extraction solvent (mixture of ethanol and water, in volume ratio, ethanol:water=80:20)

[0056] During the extraction, the centrifuge tube containing the extraction solvent was placed in an ultrasonic cell breaker (Sonics, USA) for ultrasonic extraction for 1 h.

[0057] (2) Prepare the liquid to be tested

Abstract

The application belongs to the technical field of tobacco metabolomics and particularly relates to a method for detecting polyphenolic compounds in tobacco leaves. The method can simultaneously detectone or more of kaempferol, anthocyanin, delphinidin chloride, rhamnazin, chlorogenic acid, delphinidin glucoside, procyanidine B2, anthocyanin rhamnoside, rutin and scopoletin in tobacco leaves. Themethod comprises: extracting pretreated fresh tobacco leaves to obtain a liquid to be detected and carrying out liquid chromatography-tandem mass spectrometry detection and analysis. Compared with theconventional polyphenol detection and analysis method, the detection method has the advantages of high analysis speed, many research objects, good precision, high adding standard recovery rate and good stability, and has a good practical value and a promotion and application significance.

Description

technical field [0001] This application belongs to the technical field of tobacco metabolomics, and specifically relates to a patent application for a detection method of polyphenolic compounds in tobacco leaves. Background technique [0002] my country is the largest tobacco producer in the world. As a product with high tax rate, tobacco leaves are directly related to the development of our national economy in terms of output and quality. Tobacco components are relatively complex, among which plant polyphenols play a vital role in improving the yield and maintaining the quality of tobacco leaves. [0003] Polyphenols are an important class of secondary metabolites in the process of plant growth and development. Although there are many kinds of polyphenols, statistics show that among them, rutin and chlorogenic acid have the highest proportion, and the proportion of chlorogenic acid in the total polyphenols can even reach 75% to 90%. As far as the detection of polyphenols ...
