The invention discloses a method for separating and preparing five high-purity anthocyanidin monomers from grape skins, relating to the preparation of natural pigments and belonging to the fields of food safety and food analytic chemistry. The method comprises the following steps of: firstly, carrying out anthocyanidin digestion on carbernet sauvignon grape skins to obtain crude extractive of anthocyanidin; then purifying the crude extractive by using XAD-7HP macroporous resin, and removing most of moisture absorption components such as saccharides, inorganic salts, phlegmatic temperament and the like in the crude extractive; and carrying out preparative liquid phase separation on the extractive of anthocyanidin subjected to the preliminary purification to obtain the high-purity monomers of the anthocyanidin. The method has the advantages of only using alcohol and acid as a solvent in stages of extraction, purification and separation in the preparation of the anthocyanidin and being environment-friendly, simple, convenient and feasible. The five anthocyanidin monomers obtained after purification are respectively delphinidin glucoside, 3'-methyl delphinidin glucoside, dimethyl delphinidin glucoside, dimethyl delphinidin acetylation glucoside, and dimethyl delphinidin trans-tonka-bean acetylation glucoside, the purities of which are sequentially 99.9%, 99.9%, 99.9%, 99.9%, 91.7%, 95.5%.

The invention relates to an anthocyanin synthesis related protein IbMYB113 as well as a coding gene and application thereof. The amino acid sequence of the IbMYB113 protein is as shown in SEQ ID NO: 2; the nucleotide sequence of the coding gene is shown as SEQ ID NO: 1. According to the invention, IbMYB113 protein and a coding gene thereof are cloned from red sweet potato peel, the IbMYB113 gene is connected to a plant expression vector, a transgenic tobacco plant over-expressing IbMYB113 is obtained through agrobacterium-mediated genetic transformation, pigment accumulation of the whole plant is shown, and the content of delphinidin and cornflower pigment is detected to be obviously increased by high performance liquid chromatography. The IbMYB113 protein and the coding gene thereof provided by the invention have important theoretical significance and application value for promoting synthesis and accumulation of plant anthocyanin.

According to the method, the blackcurrant waste residue produced from industrial production is taken as a raw material, and homogenized pulverized coupling enzyme hydrolysis technology, negative-pressure cavitation reinforced extraction technology, macroporous adsorption resin concentration technology, medium-pressure quick-flash reverse chromatographic separation technology and low-temperature crystallization and recrystallization technology are adopted to obtain four anthocyanin monomer ingredients including delphinidin 3-O-glucoside, delphinidin 3-O-rutinoside, cyanidin 3-O-glucoside, and cyanidin 3-O-rutinoside and having the purity reach 95%. Provided is a method for producing lilies containing a blue pigment (delphinidin) in the petals thereof by introducing a foreign gene into lilies. The method pertaining to the present invention is a method for producing lilies containing delphinidin in the petals thereof, including the following steps: introducing, into a lily, a F3'5'H gene derived from a campanula and comprising a nucleotide sequence encoding a peptide having flavonoid 3'5'-hydroxylase (F3'5'H) activity, such as a nucleotide sequence represented by SEQ ID NO: 1 or SEQ ID NO: 11; while also introducing a F3'H gene fragment derived from a lily and comprising a nucleotide sequence encoding a peptide having flavonoid 3'-hydroxylase activity, such as a nucleotide sequence represented by SEQ ID NO: 3 or SEQ ID NO: 16; and inhibiting the expression of endogenous F3'H expression, which acts on cyanidin synthesis in lily petals, while the F3'5'H gene that has been introduced acts to cause the synthesis of delphinidin.
