It is obtained from cruciferous vegetables such as broccoli, Brussels sprouts, and cabbages. It is produced when the enzyme myrosinase transforms glucoraphanin, a glucosinolate, into sulforaphane upon damage to the plant (such as from chewing), which allows the two compounds to mix and react. Extracting and purifying method of high-purity sulforaphane

The invention discloses a method for extracting high-purity sulforaphane from broccoli seeds; and the method comprises the following steps of: (1) after crushing dry broccoli seeds, adding water to the dry broccoli seeds to hydrolyze the broccoli seeds, so as to obtain a solid-liquid mixture; (2) adding anhydrous ethanol to the mixture; ultrasonically extracting the mixture; and carrying out suction filtration or double-layer gauze filtering on the mixture; (3) concentrating and evaporating ethanol from a filter liquor in a vacuum state; (4) carrying out chromatography on a material liquor by using a macroporous resin column and eluting the material liquor by using an ethanol solution in a gradient way; (5) collecting an eluting solution containing sulforaphane; carrying out chromatography on the eluting solution by using an alumina column; and concentrating a sulforaphane chromatography solution to form an extract so as to obtain a crude sulforaphane product; (6) adding water to the obtained crude product so as to dissolve the crude product; and extracting the dissolved crude product by using a polar solvent; and (7) collecting an organic layer; and concentrating the organic layer in a vacuum state so as to obtain an oily extract, so that the high-purity sulforaphane is obtained. As the high-purity sulforaphane can be directly extracted from the broccoli seeds and broccoli related materials, the method for extracting the high-purity sulforaphane from the broccoli seeds, disclosed by the invention, has the advantages of simple process, stable product, simple equipment and safety in production and is suitable for enterprises for industrially extracting the sulforaphane.

The invention relates to a method for extracting sulforaphane from broccoli. The method comprises the following steps: (1) drying edible flowering branches of fresh broccoli to constant weight and crushing to obtain broccoli dry powder; (2) carrying out vacuum filtration on newly harvested crushed mustard seeds to remove impurities to obtain a crude enzyme; (3) adding the crude enzyme into the broccoli dry powder, uniformly stirring, and carrying out enzymolysis and ultrasonic vibration to extract to obtain extracting liquor which is screened and deslagged to obtain a filtrate; carrying out vacuum filtration on the filtrate to obtain sulforaphane coarse extracting liquor; (4) vacuum concentrating the sulforaphane coarse extracting liquor to obtain sulforaphane coarse extracting aqueous liquor; (5) adding ethyl acetate into the sulforaphane coarse extracting aqueous liquor to extract to obtain an ethyl acetate phase, carrying out vacuum concentration on the ethyl acetate phase to get paste, and then eluting the paste and further vacuum-concentrating to obtain a paste sulforaphane coarse extract; (6) freezing and drying the paste sulforaphane coarse extract to constant weight to obtain the sulforaphane coarse product which is 1-5% in purity. The method provided by the invention is simple, efficient and low in cost.

Method for preparing docetaxel and sulforaphane loaded self-assembled nano-particle and application of nano-particle

The invention relates to the technical field of medicines, and provides a docetaxel and sulforaphane loaded hyaluronic acid modified poly(lactic-co-glycolic acid) copolymer self-assembled nano-particle, a preparation method thereof and an application of the self-assembled nano-particle in preparing a medicine for treating breast cancer. The microscopic structure of the nano-particle comprises a hydrophobic PLGA (poly(lactic-co-glycolic acid)) core and a hydrophilic HA (hyaluronic acid) shell, wherein the PLGA core is used for wrapping a medicine DTX (docetaxel) or SFN (sulforaphane), and the hydrophilic shell HA can target a breast cancer stem cell highly expressing a CD44 receptor. In-vitro cytotoxicity tests indicate that the drug-loaded nano-particle can play a good role in resisting differentiation of mammary gland cells and breast cancer stem cells when compared with free medicines; in-vivo tumor inhibition experiments indicate that the drug-loaded medicine is more effective than free medicines and combined free medicines, and has a small systematic toxicity. The nano-particle can play a role in simultaneously performing target differentiating on mammary gland cells and breast cancer stem cells, and a new strategy is provided to treatment of breast cancer.

The invention discloses a chemical synthesis method of sulforaphane. The method is characterized by comprising the following steps of: (a) in the presence of a catalyst, heating 1,3-propylene glycol and hydrobromic acid for reacting to obtain 3-bromine-1-propyl alcohol; (b) reacting dimethyl sulfoxide and sodium hydride at normal temperature, after the dimethyl sulfoxide and the sodium hydride completely react, adding the 3-bromine-1-propyl alcohol, and stirring; after the 3-bromine-1-propyl alcohol completely reacts, washing with a small quantity of water; extracting by using dichloromethane, taking an organic layer, and carrying out spin evaporating on the organic layer to obtain white solid 4-methylsulfonyl-1-butanol; (c) dissolving the 4-methylsulfonyl-1-butanol in an organic solvent, adding a chlorinated reagent for a chlorinated reaction to obtain 1-chlorine-4-methylsulfonyl butane; and (d) under the protection of nitrogen, dissolving the 1-chlorine-4-methylsulfonyl butane in an organic solvent, adding a thiocyanide reagent and heating, after the reaction ends, washing with water, extracting by using an organic solvent, and carrying out spin evaporating to remove the solvent to obtain faint yellow oily liquid, i.e. the sulforaphane. The method has the advantages of simpleness of operation, high yield and stable product.

Method for preparing sulforaphane

The invention belongs to the field of natural product separation and discloses a method for preparing sulforaphane, which comprises the following steps of: 1) crushing a radish seed medicinal material, adding water in an amount which is 2 to 3 times that of the crushed medicinal material, regulating the pH to be 4-5, performing enzymolysis at the temperature of between 45 and 50DEG C for 30 to 60 minutes, quickly heating to 80DEG C for deactivating enzymes, adding water in an amount of 5 to 6 times that of the solution, stirring, extracting, filtering out liquid, adding water in an amount which is 5 to 6 times that of the materials, extracting, mixing liquid in two times, and filtering to obtain extracting solution; 2) adsorbing the extracting solution by using macroporous resin, washing with water to reach neutrality, eluting by using 50- to 70 percent of ethanol solution in an amount which is 5 to 6 times that of the solution, collecting eluent, performing nanofiltration by using a nanofiltration membrane, and concentrating the passing liquid by using a reverse osmosis membrane to obtain concentrated solution; and 3) extracting the concentrated solution by using dichloromethane, and recovering reagents from extracting solution under reduced pressure to obtain the sulforaphane. The sulforaphane is prepared by using the macroporous resin and a membrane separation technology; and the process is easy to operate, the production cost is low and the method is suitable for industrial production.

Also the current invention relates to a composition comprising medium chain triglycerides (MCT), Epigallocatechin-3-gallate, curcumin, compositions comprising glucosinolates and / or derivatives thereof, such as sulforaphane (SFN). The invention provides that administering a composition comprising curcumin, EGCG, sulforaphane, alone or combined with a ketogenic diet or a modified ketogenic diet (low carbohydrate diet) alone or supplemented with MCT improves resistance of normal cells to cytotoxicity of cancer treatments, in turn, reducing their negative side effects. Increasing normal cells, tissues or organs' resistance to chemotherapeutic agents, the invention improves patients' tolerance to anti-cancer treatments' toxicity, which, in turn, contributes to enhance their efficacy, leading to increased survival of the subject treated with the current invention.
