Construction method of diabetic cardiomyopathy animal model

A technology of diabetic cardiomyopathy and animal models, applied in the field of pharmacology, can solve the problems of not fully reflecting the characteristics of chronic pathological and physiological changes of diabetic cardiomyopathy, high animal mortality (diabetic cardiomyopathy itself, great differences in the degree of pathological changes, etc., Achieve good practical application value, low animal mortality, and simple operation

Problems solved by technology

At present, there are few research models of diabetic cardiomyopathy, only reports of the hyperglycemia state induced by injection of streptozotocin (STZ) to establish a spontaneous model, but the modeling time is long (especially type Ⅱ diabetic cardiomyopathy), animal The mortality rate is high (diabetic cardiomyopathy itself is one of the reasons), the modeling rate of the model is low, and the degree of pathological changes varies greatly, which cannot fully reflect the chronic pathological and physiological changes in the occurrence and development of diabetic cardiomyopathy

Embodiment Construction

[0026] The present invention will be described in further detail below in conjunction with the accompanying drawings and embodiments.

[0027] 1. Experimental animals

[0028] Seventy SD rats, male, weighing 150-180 g, were purchased from the Experimental Animal Center of Air Force Military Medical University. The breeding temperature is 22-26°C, the relative humidity is 35%-70%, natural light source, free drinking water.

[0029] Two, grouping

[0030] The first group: blank control group (20 rats)

[0031] The second group: simple diabetes group (25 rats)

[0032] The third group: ISO intervention group (25 rats)

[0033] The second group and the third group respectively designed 5 more rats to prepare for the loss of the experimental process.

[0034] 3. Modeling method

[0035] After adaptive feeding, 20 of the 70 male rats were randomly selected as the first group (blank control group), and the remaining rats were classified as the experimental group. The blank con...

Abstract

The invention discloses a construction method of a diabetic cardiomyopathy animal model. An SD (Sprague Dawley) rat under high-sugar and high-fat feeding is subjected to multiple abdominal injection with STZ (streptozocin); after the diabetics diagnosis standard is reached, subcutaneous injection with isoprenaline is performed; after 2 weeks of continued feeding, left ventricular cannulation and three-lead electrocardiogram test of ventricular hemodynamic parameters and electrocardiogram parameters shows that the rat experiences cardiac dysfunction; heart weight index and left ventricular index show that the rat experiences evident ventricular hypertrophy; biochemical analysis on rat blood shows that the rat has evident myocardial damage; HE (hematoxylin-eosin) pathological staining satisfies pathological changes in diabetic cardiomyopathy; Masson collagen staining satisfies myocardial connective tissue proliferation of diabetic cardiomyopathy. The construction method has the advantages of good operational simplicity, short construction time, low mortality in construction phase, high construction success rate and the like, and the pathological process of myocardial damage due to human type 2 diabetes mellitus can be well simulated.

Description

technical field [0001] The invention belongs to the field of pharmacology, and in particular relates to a method for making a rat type II diabetic cardiomyopathy model. Background technique [0002] Diabetes mellitus (DM) is a metabolic disease caused by insufficient insulin secretion or peripheral tissue insensitivity to insulin, characterized by persistent hyperglycemia, and may cause various tissues and organs (such as eyes, kidneys, , heart, blood vessels, nerves, etc.) long-term damage, insufficiency or failure, most of which are type 2 diabetes. According to statistics, in 2015, about 415 million people aged 20 to 79 in the world suffered from diabetes (8.8% prevalence rate), and another 318 million people had impaired glucose tolerance (pre-prevalence rate was 6.7%). China is the country with the largest number of diabetic patients in the world. In 2015, the number of patients reached 109.6 million, and 1.3 million people died of diabetes and its complications. At t...
