Problems solved by technology

[0007] 3) Since the maxillary second molars are located deep in the mouse mouth, it is difficult to study other models that use periodontitis as an influencing factor. For example, when studying the effect of periodontitis microenvironment on immediate implant surgery on osseointegration, The anatomical position of the maxillary second molar is relatively deep. Even if the periodontitis model is successfully established, the micro-implants cannot be implanted in this position, which brings difficulties to the later composite model research

Embodiment 1

[0028] 1. Materials

[0029] Twelve 8-week-old C57 / BL6 mice, weighing 24-26 g, were provided by Nanjing Southern Model Animals and were raised in an SPF-grade environment in the Animal Center of our hospital. This experiment was approved by the Ethics Committee of our hospital. The non-absorbable suture woven with 5-0 silk thread was provided by Johnson & Johnson, and the model was Ethicom Mousse SA82G. Micro-CT is provided by Scanco Medical Company in Switzerland, the model is micro-CT 50.

[0030] 2. Experimental method

[0031] 1. Establishment of mouse periodontitis model based on the mandibular first molar:

[0032] After 12 C57 / BL6 mice were fed adaptively for one week, the mice were intraperitoneally anesthetized with 1.5% pentobarbital sodium at a dose of 40 mg / kg;

[0033] The mouse was fixed on the mouse platform in lying position, the head was cushioned, and the mouth of the mouse was stably opened with a gag, exposing the mandibular molar area. Clean the mandib...

Abstract

The invention discloses a method for establishing a mouse periodontal disease model. The method comprises the following steps of (1) performing anaesthesia on the abdomen of a mouse to be subjected to model establishing, and fixing the mouse on a mouse table in a lying manner; (2) cushioning the head of the mouse, enabling the mouse to stably open the mouth with a mouth gag to expose a mandibular molar region, then cleaning a mandibular molar and cheek and tongue side gum with physiological saline, and sufficiently drying a surgery region with a dry cotton ball; (3) inlaying suture lines into a position under the contact point of adjacent surfaces of a first molar and a second molar of the left-side lower jaw with microscopic needle forceps, and after inlaying the suture line is guaranteed, tying a knot at the position of the mesial tooth neck part of the first molar of the lower jaw; and (4) inspecting the stability degree of the knotted suture lines, and after the stability degree is confirmed, completing establishing of the periodontal disease model. According to the method disclosed by the invention, the method for establishing a novel mouse periodontitis model is realized based on the first molar of the lower jaw. The imageology analysis result on local tissue of established mouse model certificates that by the method, the mouse periodontitis model can be successfully established, and the pathology process of a clinical periodontal disease is fully simulated.

Description

technical field [0001] The invention belongs to the technical field of periodontology, and in particular relates to a method for establishing a mouse periodontal disease model. Background technique [0002] Periodontal disease is a common chronic infectious oral disease, and its direct trigger is the plaque biofilm in the gingival sulcus. As the main disease leading to tooth loss, periodontal disease manifests as chronic progressive periodontal tissue destruction characterized by alveolar bone resorption. The results of epidemiology show that the prevalence of periodontal disease in my country's children and adolescents is as high as 90%, and the prevalence of periodontal disease in adults can be as high as 97%. However, the prevalence of periodontal disease has seriously affected the masticatory function and quality of life of Chinese adults. Therefore, it is necessary to use model animals to further study the mechanism of periodontal disease. [0003] As a representative ...
