Problems solved by technology

At present, there have been some relevant studies and reports on EHDV species or serogroup-specific PCR detection; however, these detections require special instruments and equipment, and the detection process is relatively complicated, which is difficult to apply to on-site detection
EHD is an exotic disease. Although there has been no report of the disease being found in my country, with the continuous increase in the trade volume of imported animals and animal products in China, there is a risk of EHD being introduced into my country. Once introduced, it will affect the breeding of our country. Therefore, it is urgent to establish a better EHDV detection method suitable for on-site rapid detection to prevent the introduction or spread of EHDV

Embodiment

[0041] 1. Materials and methods

[0042] 1. Plasmid and nucleic acid samples for testing

[0043] In this example, according to the conserved non-structural protein NS1 gene (Segment 5) of deer epidemic hemorrhagic fever virus published in NCBI GenBank, the gene fragment was synthesized by Shenggong Bioengineering (Shanghai) Co., Ltd. and cloned into the pUC vector, named as EHDV-Seg5. The inactivated antigen nucleic acid used in this example includes EHDV-1 and EHDV-1, bluetongue virus nucleic acid, bovine viral diarrhea virus nucleic acid, foot-and-mouth disease virus nucleic acid.

[0044] 2. Main reagents and instruments

[0045] TwistAmp exo kit (TwistDx), MMLV Reverse Transcriptase (Takara), Recombinant RNase Inhibitor (Takara), constant temperature amplification instrument (Axxin), 5415R high-speed centrifuge (Eppendorf), 7500 Fast fluorescence PCR instrument (ABI), ultra-trace nucleic acid Protein concentration analyzer (BioDrop), etc.

Abstract

The application discloses a reagent, a detection method and an application for detecting deer epidemic hemorrhagic fever virus. The reagent comprises a specific primer pair and a probe, wherein upstream and downstream primers of the primer pair are shown in SEQ ID No. 1 and 2 respectively, and the probe is shown in SEQ ID No. 3; and in the sequence shown in SEQ ID No. 3, a 32nd base modifies fluorescence quenching group-dT, a 33rd base is replaced by base analog, a 35th base modifies fluorescence group-dT, and a 3' end modifies C3Spacer. The reagent can sensitively, specifically and efficiently detect deer epidemic hemorrhagic fever virus through one-step reverse transcription recombinase polymerase amplification. By adopting the reagent, on-site detection of deer epidemic hemorrhagic fever virus can be conveniently carried out, so that the detection result can be quickly obtained; and the regent is of great significance for the rapid prevention and control of deer epidemic hemorrhagic fever virus, preventing the spread of the epidemic, and ensuring production safety to the greatest extent.

Description

technical field [0001] The application relates to the field of detection of deer epidemic hemorrhagic fever virus, in particular to a reagent, detection method and application for detection of deer epidemic hemorrhagic fever virus. Background technique [0002] Epizootic hemorrhagic disease of deer (EHD) is a disease caused by deer epidemic hemorrhagic fever virus (EHDV) of the Reoviridae family Orbivirus genus. EHD is a non-contact infectious disease transmitted by the insect Culicoides, affecting wild and domestic ruminants, and is one of the important vector-borne diseases. Under natural conditions, EHDV can cause infection of livestock such as cattle and sheep, as well as domesticated and wild ruminants such as white-tailed deer, mink, and eland antelope, among which white-tailed deer is the species most affected by EHDV. After deer are infected with the virus, their body temperature rises, the mucous membrane and serosal surface hemorrhage extensively, and the mortalit...
