Embodiment 1

[0088] Embodiment 1. The nematode lifespan experiment of verapamil

[0089] 1.1 Method

[0090] Verapamil hydrochloride (CAS: 152-11-4) was purchased from Shanghai Titan Technology Co., Ltd.; wild-type Caenorhabditis elegans (N2) was purchased from CAENORHABDITIS GENETICS CENTER (CGC).

[0091] Transfer the synchronized nematodes to the nematode growth medium (NGM) of the blank control group and the verapamil administration group (100 μM, 400 μM) at the L4 stage, with about 80-90 nematodes in each group, and change every 3 days Primary medium, cultured at 20°C. The number of surviving nematodes was recorded every day, and the dead nematodes were removed from the culture dish until all the nematodes died. Summarize the data, draw the survival curve with GraphPad, and calculate the significant difference (Log-rank test). Three valid replicate experiments were performed.

[0092] 1.2 Results

[0093] The life experiment data are shown in Table 1 and figure 1 shown.

Embodiment 2

[0099] Example 2. Verapamil improves healthy physiological parameters of nematodes

[0100] 2.1 Method

[0101] To further evaluate the effect of verapamil on nematode health parameters, the locomotion behavior, body swing ability, vulvar integrity defect and osmotic stress capacity of the nematode were tested.

[0102] Nematode moving behavior experiment: Cultivate nematodes according to the method of nematode lifespan experiment, pick 20 to 30 nematodes in the control group and verapamil group respectively on the 2nd, 4th, 6th, 8th, 10th, 12th, and 14th days of adult nematodes , respectively record the crawling trajectory of each nematode within 30 seconds, and process it with ImageJ to obtain the crawling speed of the nematode. After the experiment, statistical data were calculated using Graph Pad to calculate significant differences (Two-way ANOVA, Sidak multiple comparisons test), and the error bars indicate the standard error of the mean (SEM). Three valid replicate expe...

Embodiment 3

[0113] Example 3. Verapamil reduces cell senescence rate

[0114] The vast majority of normal cells are considered to have limited ability to divide, and enter the senescent state after being unable to divide. Senescent cells usually become larger in size and have highly active β-galactosidase (β-galactosidase), which can be used for cell senescence β-galactosidase staining kit (Beiyuntian) was used to stain β-gal to evaluate the degree of cell senescence.

[0115] In this embodiment, SA-β-Gal was used as a senescent cell marker, and it was stained with chemical reagent X-Gal to make it blue-green.

Abstract

The invention provides an ageing resisting purpose of verapamil, and specifically provides a purpose of the verapamil, or an optical isomer thereof, or pharmaceutically acceptable salt thereof to preparation of ageing resisting compositions or preparations. The experiment indicates that the verapamil can significantly prolong the life of threadworms; the threadworm health parameter evaluation experiment confirms that the verapamil can improve the movement behavior capacity, the body swinging capacity and the osmotic pressure stress capacity of the threadworms, and reduce vulva integrity defects; and ageing cell dyeing experiment indicates that the verapamil can reduce the ageing rate of human embryonic lung cells. Therefore, the verapamil can be applied to respects of prolonging life, delaying ageing, improving health physiological parameters, reducing the incidence rate of ageing related diseases and the like. The invention also provides products of ageing resisting medicines, health-care products, cosmetics and the like.

Description

technical field [0001] The invention relates to the field of medicine, in particular to the anti-aging application of verapamil. Background technique [0002] Aging is a complex and changeable gradual process, which is affected by various mechanisms. With the continuous growth of age, the functions of various tissues and organs of the body begin to be damaged, and the metabolism and stress response ability decline, often accompanied by diabetes, tumors, cardiovascular disease, etc. diseases, neurodegenerative diseases, etc. Aging and aging-related diseases seriously threaten human health and quality of life, and increase the social medical burden. Although aging is irreversible, it is possible to delay aging. It is the most effective intervention strategy to delay aging, prolong healthy lifespan, and reduce the incidence of aging-related diseases through anti-aging drugs. [0003] Anti-aging drugs are developed with the aim of improving life efficiency, ie survival time an...
