According to the invention, for illumination in each polarization modulation direction, two sinusoidal structured light illumination patterns with complementary phases are collected and the two sinusoidal structured light illumination patterns are averaged to obtain a uniform light illumination pattern; the uniform light illumination pattern is combined with one of the sinusoidal structured light illumination patterns, and an optical tomographic image is solved by using a HiLo algorithm; only the three structured light patterns need to be averaged to obtain a uniform light illumination pattern, and then the uniform light illumination pattern is combined with one of the structured light illumination patterns, so that the optical tomography pattern can be solved by utilizing a HiLo algorithm. According to the invention, a detection end light splitting and collection method is adopted, and extra collection time does not need to be increased; the system and method have great advantages in the aspects of imaging speed, less photobleaching and the like.
