Embodiment

[0037] This example provides a method for analyzing the tissue distribution of secondary metabolites in geckos, including the following steps:

[0038] S1: Cut the cross-section of the hook kiss tissue, make a frozen section, and obtain a complete tissue sample imprint on the glass slide;

[0039] S2: Analyze the imprint of the tissue sample obtained in step S1 by a desorption electrospray ionization mass spectrometer to obtain a mass spectrometry imaging file;

[0040] S3: Process the mass spectrometry imaging file obtained in step S2 by image processing software, and obtain a visualized picture of the distribution of secondary metabolites in the tissue of the gecko.

[0041] In step S1, the method for making frozen sections is specifically:

[0042] Cut the cross-section of the glyphus tissue, which includes the roots, stems, leaves and buds of the glyphs, soak and fix the roots, stems, leaves, and buds of the glyphs in fresh tissue fixative solution, and fix them for 24 ho...

Abstract

The invention provides a method for analyzing the distribution of secondary metabolites in gelsemium elegans in a tissue, which comprises the following steps: shearing the cross section of a gelsemiumelegans tissue, making a frozen section, obtaining a complete tissue sample print on a glass slide, and analyzing the tissue sample print obtained in the step S1 by using a desorption electrospray ionization mass spectrometer to obtain a mass spectrum imaging file, and finally processing the mass spectrum imaging file obtained in the step S2 through image processing software to obtain a visual picture of the distribution of the secondary metabolites in the gelsemium elegans in the tissue. According to the method disclosed by the invention, the problem of high sample consumption in a pretreatment process is solved, the frozen section is combined with a mass spectrum imaging technology, and finally, a mass spectrum imaging file is processed by image processing software, so that as many as 26 clear pictures of the spatial distribution of gelsemium elegans secondary metabolites in gelsemium elegans roots, stems, leaves and buds can be obtained; and the analysis and detection process is simple and convenient.

Description

technical field [0001] The invention belongs to the technical field of analysis methods for chemical components, and in particular relates to a method for analyzing the distribution of secondary metabolites in tissues of geckos. Background technique [0002] Ginkgo, also known as heartbroken grass, kudzu, etc., is a world-renowned highly poisonous plant. There are three species in the world, namely: Gelsemium elegans Benth, mainly distributed in Fujian, Zhejiang, Yunnan, Guangxi, Hunan, Guizhou, Jiangxi and other southern regions and Southeast Asia; (L.) J.st-Hil) and marsh jasmine (Gelsemium rankinii Small.), mainly distributed in North American countries. Chinese medicine has long recorded the hook kiss, as described in the "Dictionary of Chinese Medicine": its roots, ridges, and leaves can be used as medicine, warm in nature, pungent, bitter in taste, and highly poisonous. It has the functions of expelling wind and attacking poison, dispelling stagnation, and relieving ...
