Embodiment 1
[0093] In vitro test against liver cancer cells
[0094] Method: use 3 [H]thymidine incorporation assay to investigate the effect of clofazimine on cell proliferation. Adherent cells (5 / 000-10 / 000) were plated into 24-well Corning tissue culture dishes and exposed to clofazimine containing vehicle or different concentrations (10 -9 -10 -5 mol / L) medium (MEM 5% FBS). For Novikoff, which is a detached rat cell line, 2000 cells were suspended in 2 ml DMEM (5% FBS) and maintained under the same conditions as for adherent cells. The medium was replaced with fresh medium every other day. At the end of the treatment period (5-10 days), by adding 0.5 μCi to each well [ 3 H] thymidine and a final incubation of 4 hours to measure thymidine incorporation into the cell culture. The amount of incorporated radioactivity was determined using a β-scintillation counter (18). Results are expressed as actual counts per minute - drug concentration used in medium or % inhibition (reduction ...
Embodiment 2
[0100] In vitro test of anti-HepG2 cells
[0101] HepG2 cells plated in 6-well plates were treated with clofazimine (0-5 μM) for 1, 3 or 7 days, and the remaining viable cells were counted using the trypan blue dye method. All counts were obtained in quadruplicate. The results are attached figure 2 , where the values represent the mean ± standard error.
Embodiment 3
[0103] In Vitro Test of Novikoff Against Rat Hepatoma Cells
[0104] Rat hepatoma cells novikoff were grown in test tubes and treated with different concentrations of clofazimine (0-10 μM) for 1, 3 or 7 days. At the end of the treatment period, the number of remaining viable cells was counted using the trypan blue method. All counts were obtained in triplicate and values represent mean ± standard error. The results are attached image 3 shown.
