Problems solved by technology

[0005] Judging from the current research situation, most of the fermentation of traditional Chinese medicine still adopts solid fermentation or natural fermentation, which not only has complex bacterial strains, but also has a long transformation cycle, which brings great difficulties to the directional transformation of key components in traditional Chinese medicine. There is no report on solid or liquid microbial transformation and bioavailability of Scrophulariaceae

Embodiment 1~3

[0024] Experiments on Transformation Effects of Different Strains of Scrophulariaceae

[0025] See attached figure 1 Scrophulariae scrophulariae was transformed using alcoholic yeast, Meishan yeast and brewer's yeast as strains, and harpagoside was extracted and determined for untransformed and transformed Scrophulariae scrophulariae. The conditions and results were as follows:

[0026] Each strain was stored on a slant in wort solid medium, activated at 28°C for 24 hours, and then inoculated on wort liquid medium (no agar, pH 5.5). The liquid seeds were cultured in a 250 ml shaker bottle with 50 ml liquid, the fermentation temperature was 28° C., the shaker speed was 150 r / min, and cultured for 24 hours to obtain a culture solution.

[0027] Accurately weigh 2 grams of Scrophulariaceae that has been crushed to 20-60 meshes, dissolve it in 50 ml of distilled water, adjust the pH to 5.5, and sterilize at 121°C for 15 minutes to obtain Scrophulariaceae culture medium; Insert 1...

[0033] Experiments on Transformation Effects of Scrophulariaceae with Different pH

[0034] According to the method of Example 1, only Scrophulariae cerevisiae was used as the strain to transform Scrophulariaceae culture medium with different pH, and the transformed Scrophulariaceae was extracted and its content was determined. The results are shown in Table 2:

[0035] Table 2

[0036]

[0037] The comparative analysis in Table 2 found that the use of different pH mediums of Scrophulariae scrophulariae significantly affected the content of harpagoside in Scrophulariae scrophulariae, and the yield of harpagoside was the highest at pH=5.5.

Embodiment 7~10

[0039] Experiments on Transformation Effects of Different Inoculation Ratio on Scrophulariaceae Scrophulariae

[0040]The cells were obtained by culturing according to the method of Example 1. Saccharomyces cerevisiae was used as the transformed microorganism, and the culture solution was inserted into the culture medium of Scrophulariaceae Scrophulariae according to different inoculation ratios for transformation. The content of harpagoside was determined for the transformed Scrophulariaceae Scrophulariae. The results are shown in table 3:

[0041] table 3

[0042]

[0043] Note: Each ml of culture medium contains 1×10 strain cells 8 cfu.

[0044] It can be seen from Table 3 that the inoculation ratio of 1:1 is very effective for the transformation of Scrophulariaceae, and the yield of harpagoside is significantly higher than that of other groups. Therefore, it is more beneficial to select the inoculation ratio of 1:1 for the transformation of Scrophulariaceae. Extracti...

Abstract

The invention discloses a method for improving the extraction ratio of arpagoside in radix scrophulariae by fermentation. The method comprises the following steps: (1) saccharomyces cerevisiae is cultured on beerwort solid culture medium and transferred to beerwort liquid culture medium for culturing so as to obtain culture solution; (2) the radix scrophulariae is mixed with distilled water, and the mixture is subjected to sterilization treatment to obtain radix scrophulariae culture medium; (3) the culture solution is injected in the radix scrophulariae culture medium to culture and convert the radix scrophulariae so as to obtain converted radix scrophulariae culture solution; and (4) the converted radix scrophulariae culture solution is centrifuged to remove residue so as to obtain supernatant, and the arpagoside in the supernatant is extracted by the solvent extraction method. In the method, the microbe fermentation method is adopted to make the bioconversion of the radix scrophulariae, so that compared with the unconverted radix scrophulariae, the arpagoside content is improved by 30 to 50 percent.

Description

technical field [0001] The invention relates to the field of bioengineering, in particular to a method for improving the extraction rate of harpagoside, the main functional component of Scrophulariae scrophulariae, by using a microbial fermentation method. Background technique [0002] my country's traditional Chinese medicine has a long history and rich resources of traditional Chinese medicine. It plays a huge role in ensuring the health of the people. The international community is increasingly aware of the important role of traditional Chinese medicine in the treatment and prevention of diseases. However, in recent decades, my country has made slow progress in the modernization of traditional Chinese medicine. The main reason is that the processing technology of traditional Chinese medicine is outdated and the active ingredients in traditional Chinese medicine are not clear. In recent years, the microbial fermentation technology of traditional Chinese medicine, which is ...
