Problems solved by technology

The currently reported quantitative analysis method for the simultaneous determination of free and bound sterols in tobacco is mainly gas chromatography. The shortcoming of this analytical method is that it can only detect ergosterol, campesterol, stigmasterol and β-sitosterol, but cannot be determined simultaneously. cholesterol

Embodiment Construction

[0016] Below by embodiment the invention is further elaborated:

[0017] Cholesterol with a purity greater than 95%, stigmasterol with a purity greater than 95%, β-sitosterol with a purity greater than 95%, ergosterol with a purity greater than 98%, campesterol with a purity greater than 97%, and 5a with a purity greater than 97% - The solution formed by dissolving cholestane in dichloromethane was configured into 5 standard solution samples (standard solution 1 to standard solution 5) of sterols (including internal standard) with different concentrations, and the corresponding concentrations were 16, 32 , 64, 128, and 256 μg / mL, and the internal standard concentration was 10 μg / mL. Take 200 μL of the above standard solution and filter it through a 0.45 μm filter membrane, dry it with nitrogen, add 200 μL bis(trimethylsilyl)trifluoroacetamide (containing 1% TMCS), heat at 80°C for 60 min, take 1 μL for GC — MS (gas chromatography analyzer) analysis. The analysis conditions a...

Abstract

The invention discloses a method for measuring sterol in tobaccos. The method comprises the following steps of: (1) deploying sterol standard templates containing internal standard with different concentrations, carrying out GC-MS (Gas Chromatography-Mass Spectrometer) analysis after derivatization, and plotting by utilizing the ratio of the sterol to internal standard peak area and the ratio of the sterol to internal standard concentration to obtain a standard curve; (2) carrying out acid hydrolysis, saponification and direct ultrasonic extraction on samples to be measured, and taking 1 muL for carrying out GC-MS analysis after derivatization; and (3) reading the ratio of the sterol to the internal standard concentration from the standard curve by utilizing the ratio of the sterol to theinternal standard peak area, and then calculating the amount of the sterol. The invention can be used for analyzing cholesterol, ergosterol, campesterol, stigmasterol and beta-sitosterol with a plurality of forms in tobacco leaves at the same time, has the advantages of accurate measurement and simple and convenient operation, and can be widely applied to tobacco industry.

Description

technical field [0001] The present invention relates to the determination method of sterol, especially the determination method of sterol in tobacco. Background technique [0002] Sterols are a class of important compounds in tobacco. They are considered to be the precursors of condensed aromatic hydrocarbons in cigarette smoke. They have a great impact on the quality and safety of tobacco. A comprehensive understanding of the content and form of sterols in tobacco can reduce the harm of cigarettes. Focus reduction has certain guiding significance. Sterols account for about 0.1-0.3% of the weight of tobacco, mainly including campesterol, stigmasterol, cholesterol and β-sitosterol, and moldy tobacco leaves also contain ergosterol. The first four sterols mainly exist in the forms of esters, glycosides, acyl sugar compounds and free forms. The currently reported quantitative analysis method for the simultaneous determination of free and bound sterols in tobacco is mainly gas ...
