Embodiment Construction

[0028] The invention detects the single nucleotide polymorphism of the mutation at the 2359th position of the Angptl6 gene of cattle, so as to be used for marker-assisted selection of growth traits of Chinese cattle cattle, and quickly establish cattle populations with excellent genetic resources. In the following, the present invention will be further described in detail in combination with the detection of specific samples and the association of traits, which is an explanation of the present invention rather than a limitation.

Abstract

The invention discloses a method for detecting single nucleotide polymorphism (SNP) of cattle Angptl6 gene. According to the invention, cattle genomewide DNA comprising Angptl6 gene is adopted as a template, and a primer pair P (comprising P1 and P2) is adopted as a primer. A PCR method is adopted for amplifying the cattle Angptl6 gene, and a PCR amplification product is digested by restrictive endonuclease ScaI. A segment after digestion is processed through agarose gel electrophoresis, and the SNP of the 2359 site on the cattle Angptl6 gene (GenBank Accession number: NW_003104015) is identified according to the results from the agarose gel electrophoresis. Because Angptl6 gene has important biological functions towards growth properties, the detection method provided by the present invention contributes to the establishing of a basis for constructing a relation between SNP of Angptl6 gene and growth properties. Also, the inventor discovers that, SNP of Angptl6 gene has a substantialinfluence on body length indexes. Therefore, the detection method provided by the present invention can be applied in the molecular marker auxiliary selection of Chinese beef selection and growth property selection, and cattle population with excellent heredity resource can be rapidly established.

Description

technical field [0001] The invention belongs to the field of molecular genetics, relates to the detection of gene single nucleotide polymorphism (Single Nucleotide Polymorphism, SNP), in particular to a method for detecting the 2359th single nucleotide polymorphism of cattle Angptl6 gene. Background technique [0002] Gene polymorphism refers to the difference in genome sequence between different species or different individuals within the same species. These differences are caused by nucleotide changes in DNA alleles in chromosomes, mainly including base substitutions, insertions, deletions, and repetitive sequences. Copy number variation. SNP is a type of genetic marker system proposed by Lander (1996), a scholar at the Human Genome Research Center of the Massachusetts Institute of Technology, which refers to the change of a single nucleotide (A / T / C / G) in the genomic DNA sequence caused by polymorphism. Its variant forms include: transversion, conversion, insertion and d...
