Embodiment 1

[0025] Embodiment 1, the effect of patulin on the growth of Pichia membranaefaciens IMI 382465

[0026] 1. Activation of Pichia membraneaefaciens IMI 382465 strain

[0027] The Pichia membranaefaciens IMI 382465 stored at -20°C was streaked on the NYDA plate. After culturing at 26°C for 72 hours, pick yeast cells from the single colony of Pichia membraneefaciens (Pichia membraneefaciens) IMI 382465 that grew well and inoculate them into NYDB liquid medium for shaking culture for 24 hours at a culture temperature of 26°C on a shaking table The speed is 200 rpm.

[0028] 2. Preparation of patulin standard stock solution

[0029] The patulin is a standard product of patulin, purchased from Qingdao Shenggong Company, and prepared into a mother solution with a concentration of 1 mg / ml when used. The preparation method is as follows: accurately weigh 10 mg of patulin standard substance, dissolve it in 10 ml of distilled water, fully stir to dissolve, and use a 0.22 μm microporous...

Embodiment 2

[0038] Example 2, Detection of the scavenging ability of Pichia membraneaefaciens IMI 382465 to patulin

[0039] 1. Activation of strains

[0040] With embodiment 1 step 1.

[0041] 2. Preparation of patulin standard stock solution

[0042] With embodiment 1 step 2.

[0043] 3. Preparation of culture medium containing different concentrations of patulin

[0044] With embodiment 1 step 3.

[0045] 4. Analysis of the scavenging ability of Pichia membraneaefaciens IMI 382465 to patulin

[0046] Inoculate the activated Pichia membranaefaciens IMI 382465 into the culture solution prepared in step 3 containing 50 μg / ml, 100 μg / ml and 200 μg / ml patulin respectively, so that the membranes after inoculation are inoculated. The cell concentration of yeast (Pichia membraneaefaciens) IMI 382465 was 2 × 10 5 cfu / ml, the culture temperature was 26°C, the shaker speed was 200 rpm, and each treatment was repeated 3 times. At the same time, a control without Pichia membraneefaciens (Pic...

Abstract

The invention discloses application of a yeast strain to elimination of patulin. The invention provides application of Pichia membranaefaciens to elimination of patulin. Experiments prove that Pichia membranaefaciens can quickly remove high concentration patulin in culture solution; patulin residues can not be detected four days after the Pichia membranaefaciens cells with concentration being 2*105cfu / ml are inoculated into the culture solution containing 100mu g / ml of patulin to be cultured; and patulin residues can not be detected six days after the Pichia membranaefaciens cells with concentration being 2*105cfu / ml are inoculated into the culture solution containing 200mu g / ml of patulin to be cultured. Application provided by the invention can provide a new solution to removal of patulin residues in such food as fruit juice.

Description

technical field [0001] The invention relates to the application of a yeast strain in eliminating patulin, in particular to the application of Pichia membraneefaciens in eliminating patulin. Background technique [0002] Patulin is a secondary metabolite of some Penicillium and Aspergillus fungi. It has extensive and strong toxic effects on humans and animals. Acute toxicity manifests as convulsions, convulsions, subcutaneous tissue edema, pulmonary and intestinal congestion, enteritis, anuria, and death; chronic toxicity can lead to gastric ulcers, immunosuppression, teratogenicity, and potential carcinogenicity, etc., and may also affect men of fertility. Patulin is soluble in water, ethanol, acetone, ethyl acetate and chloroform, slightly soluble in ether and benzene, insoluble in petroleum ether, and chemically stable under acidic conditions. [0003] Among the fungi capable of producing patulin, some fungi are plant pathogens, and the most important one is Penicillium...
