The method comprises the steps of a, strain dividing: dividing the Crassostrea sikamea into three groups according to the left shell radial ridge number of the Crassostrea sikamea, wherein the three groups comprise a ridge-free strain without a radial ridge, a less-ridge strain with the number of the radial ridges being smaller than or equal to 5 and a multi-ridge strain with the radial ridge number being larger than 6; b, strain self multiplying: taking the 3 strain individuals in the step a as parent samples, performing offspring multiplying by group self multiplying in the strains and then performing normal cultivating; c, strain purifying; taking the strains cultivated in the step b as the parent samples, eliminating the parent sample individuals with the left shell radial ridge number which is not consistent with the strain standards, repeating the group self multiplying in the strain for a plurality of times for offspring multiplying and performing normal cultivating, and performing oriented purifying to obtain a new strain with relatively stable characters.
