Embodiment Construction

[0034] In the following examples, the method for obtaining DNA from the sample to be tested is a technique well known to those skilled in the art, for example, the traditional phenol / chloroform / isoamyl alcohol method can be used, or some commercially available DNA extraction kits can be used.

[0035] As a preferred solution, this embodiment uses the Genomic DNA Extraction Kit (spin column type) of Tieniae bacteria.

[0036] The specific experimental method of this embodiment. According to conventional conditions such as the conditions described in the Molecular Cloning Experiment Guide edited by J. Sambrook et al., or according to the conditions suggested by the manufacturers of relevant reagents and instruments.

Abstract

The invention discloses a PCR detection method and a kit for detecting pathogenic microorganisms in fruit juice. The detection method comprises the followings steps: (1) adding a certain amount of a fruit juice sample to be detected into an enrichment liquid; (2) using DNA in the sample to be detected as a template after the sample is subjected to enrichment, and performing PCR amplification on four pairs of primers of SEQ ID No. 1, SEQ ID No. 2, SEQ ID No. 3, SEQ ID No. 4, SEQ ID No. 5, SEQ ID No. 6, SEQ ID No. 7 and SEQ ID No. 8; (3) detecting whether specific amplification exists in an amplification result or not and whether corresponding peak values exists in a dissolution graph to judge whether object pathogens are in the sample or not. The invention further discloses the kit for realizing the method. According to the detection method provided by the invention, a condition that whether common foodborne pathogens exist in the juice or not can be accurately and quickly detected out, and the method has excellent reproducibility and sensitivity.

Description

technical field [0001] The invention relates to a PCR detection method and a kit for detecting food-borne pathogenic bacteria in fruit juice, belonging to the technical fields of molecular biology and nucleic acid detection. technical background [0002] Juice is one of the most commonly consumed liquid beverages in people's lives. Along with people's pursuit of healthy quality of life, fruit juice beverages have become the same beverage varieties widely accepted by consumers as traditional carbonated beverages and alcoholic beverages. Most fruit juice drinks are acidic drinks, among which kiwi fruit juice has attracted much attention due to its rich vitamin C content. [0003] Kiwi fruit juice is the juice of kiwi fruit. It is known as the "king of fruits" because of its rich vitamin C content. In addition, kiwi fruit also contains good soluble dietary fiber and SOD, a phytochemical with high antioxidant activity. [0004] Although kiwi fruit juice has a high nutritional ...
