Problems solved by technology

But the research on catalpa somatic embryogenesis has not been reported yet, so the establishment of a stable and high-frequency catalpa somatic embryo regeneration system will be useful for the biotechnology breeding of catalpa, the preservation of germplasm resources, rapid propagation, genetic transformation, and artificial control of catalpa. The study of tree growth and development provides a good experimental body, which plays a very important role

Embodiment Construction

[0038] The methods used in the following examples are conventional methods unless otherwise specified.

[0039] 1 / 2 MS basic medium: refer to the literature (Murashige T, Skoog F. A revised medium for rapid groove and bioassays with tobacco tissue cultures. Physiol. Plant , 1962, 15: 473-497) formulated with only half the macronutrients.

[0040] Such as Figure 11 Among them, the main content of the in vitro culture method of catalpa is as follows: sterilization—callus induction—callus proliferation—embryogenic callus induction and somatic embryo differentiation—plant regeneration—transplantation , and take the immature cotyledon embryo as the explant concrete implementation as follows:

[0041] 1) Sterilization of cotyledon embryo

[0042] Take the immature slender capsule of catalpa, wipe and disinfect it with 75% alcohol cotton ball for 2-3 times, enter the aseptic operation table and wipe and disinfect it with 75% alcohol cotton ball for another 2-3 times, put on steril...

Abstract

An in-vitro culture method for catalpa bungei comprises 1) inducing callus, concretely, putting catalpa bungei on a callus induction medium for inducing callus, wherein the callus induction medium is obtained by adding 6-benzyladenine (6-BA) and 2,4-dichlorophenoxyacetic acid (2,4-D) on the basis of 1 / 2 MS basal medium; 2) propagating callus, concretely putting callus on a propagation medium for propagation, wherein the propagation medium is obtained by adding 6-benzyladenine (6-BA) and alpha-naphthalene acetic acid (NAA) on the basis of 1 / 2 MS basal medium; 3) inducing embryonic callus and differentiating somatic embryo, concretely putting propagated callus on an induction differentiation medium, wherein the induction differentiation medium is obtained by adding 6-benzyladenine (6-BA) and alpha-naphthalene acetic acid (NAA) on the basis of 1 / 2 MS basal medium; and 4) regenerating a plant, concretely putting somatic embryo obtained through differentiation on a regeneration medium for inducing somatic embryo to germinate, so as to obtain a catalpa bungei regeneration plant. The method aims at expanding catalpa bungei propagation coefficient and providing conditions for germplasm preservation and genetic transformation.

Description

technical field [0001] The invention relates to an in vitro cultivation method of plants, in particular to an in vitro cultivation method of catalpa. Background technique [0002] Catalpa ( Catalpa bungei C. A. Mey) belongs to Bignoniaceae (Bignoniaceae) Catalpa ( Catalpa ) is a native tree species originating in my country, and its distribution ranges from the seashore in the east to Gansu in the west, Yunnan in the south and the Great Wall in the north. This tree species has strong adaptability and wide distribution, and is known as the "King of Wood" (Wang Wencai et al. 1990). The tree is handsome, tall and straight, with many flowers and crowns. Its flowers are bell-shaped, with red spots dotted with white corollas. Every flowering period, the flowers are full of branches. It is a famous garden ornamental tree species in the history of our country. It has been planted in royal gardens, ancient temples and temples since ancient times. At the same time, catalpa is also...
