The method comprises the following steps: (1) selecting and cultivating rootstock seedlings, wherein Populus diversifolia seed bearer with good genes is used as the source of rootstock, the seed of Populus diversifolia seed bearer is soaked and then sowed, the obtained seedlings are transferred for bed cultivation for one year, and in summer at third year, the seedlings having vigorous growth are used as rootstock seedlings; (2) screening and treatment of scions, wherein the excellent Populus diversifolia-Populus pruinosa hybrid new product is selected in a mixed growth area of Populus diversifolia and Populus pruinosa as a seed tree, the seed tree is cut with stump being leaved, the sprouting branches are cultivated and vigorous branches are used as scions; (3) grafting; (4) management; and (5) secondary grafting.
