Embodiment Construction

[0014] The present invention will be described in further detail below in conjunction with specific embodiments.

[0015] Rearing of parents and collection of sperm and eggs

[0016] After the wrinkled plate abalone is taken back, it is washed and cleaned, and it is temporarily kept in 14°C circulating sea water, and the male and female individuals are kept separately. Feed the bait. During the temporary raising period, the temperature is increased by 1°C every day, and the temperature is kept constant at 16-17°C for production. Ovulation was induced by drying in the shade and heating method, and the specific operation was as follows: select mature female shellfish with full gonads, wash them and dry them in the shade for 50 minutes, then put them into a bucket and induce oviposition in seawater at 20°C. After the eggs are produced, first filter the eggs with a 300-mesh sieve, and then use a 500-mesh sieve to collect the eggs. After dilution, observe and quantify the density...

Abstract

The invention relates to a gynogenesis inducing method of haliotis discus hannai. According to the method, an electrostimulation method is adopted; ova are transferred into an electrostimulation buffer solution for balance for 2min before electrostimulation; parameters of an electric pulse instrument is regulated; 1ml of ova are sucked with a sucker and put into an electrostimulation cup, and then electrostimulation treatment can be carried out; the electrostimulation buffer solution is a Zimmerman buffer solution which specifically comprises 0.1 mmol/L of Ca(C2H3O2)2.H2O, 1 mmol/L of K2HPO4, 0.5 mmol/L of Mg(C2H3O2)2.4H2O, 0.1 mmol/L of reduced glutathione, 1% of BSA, and 0.30 mmol/L of sucrose; the parameters of the electric pulse instrument include the field intensity of 0.88 kv/cm, the capacitance of 10[mu]F, and 3 times of electric pulses. The density of ova is 7.50x10<4> /ml during electrostimulation. The gynogenesis inducing method is simple and effective. After electrostimulation induction, the cleavage rate of the haliotis discus hannai can reach 28% or above. The embryo survival rate can reach 98% or above.

Description

technical field [0001] The invention belongs to the technical field of marine organisms, and in particular relates to a method for inducing gynogenesis of abalone wrinkled. Background technique [0002] Artificial induction of gynogenesis is conducive to the rapid establishment of pure lines, so it is of great significance to the research of genetic breeding and basic biology. In terms of marine economic shellfish, research in this field is relatively lagging behind. Currently, only about ten species of marine economic shellfish have been artificially induced to study gynogenesis. These studies mainly use various radiation or chemical methods to inactivate sperm, thereby inducing gynogenesis of eggs. The homologous sperm inactivation method commonly used in research is difficult to eliminate the interference of exogenous sperm genome, so in recent years, some scholars have tried to use heterologous sperm induction to eliminate this interference, such as studies in clams suc...
