Problems solved by technology

Otoliths, horny jaws, etc. are considered to be reliable materials for identifying the age of soft fishes. In the past, some studies tried to use the growth lines on the surface of the inner shell to identify the age of soft fishes, but it was difficult to interpret the early growth lines. and cannot be used for age determination

Embodiment 1

[0060] Embodiment 1: Utilize inner shell to identify the method for the age of soft fish

[0061] The operation steps are as follows:

[0062] 1. Extraction: carefully extract the inner shell of soft fish with a scalpel and scissors to prevent the inner shell from breaking and breaking;

[0063] 2. Cleaning: Clean the mucus and other organic matter on the surface of the inner shell after taking it out with clean water;

[0064] 3. Dehydration: the cleaned inner shell is dehydrated in absolute alcohol for 30 days;

[0065] 4. Drying: Put the dehydrated inner shell in a fume hood and let it dry naturally for about 1 week.

[0066] 5. Cutting: Take out the thoroughly dried inner shell ( figure 1 ), use surgical scissors to cut off the stalk and caudal vertebrae, and keep the stem;

[0067] 6. Embedding: According to the volume ratio of 1:1 ~ 1:1.5, the cold embedding resin solution made of acrylic powder and liquid curing agent (viscosity of 60mPas, purchased from Shenzhen Sa...

Embodiment 2

[0076] Embodiment 2: A kind of preparation method of soft fish inner shell slice

[0077] The operation steps are as follows:

[0078] (1) Cutting: Take out the thoroughly dried inner shell ( figure 1 ), use surgical scissors to cut off the stalk and caudal vertebrae, and keep the stem;

[0079] (2) Embedding: A cold embedding resin solution prepared by acrylic powder and liquid curing agent (viscosity 60mPas, purchased from Shenzhen Sanhao Instrument Equipment Co., Ltd.) at a volume ratio of 1:1 to 1:1.5, Pour into the mold so that the solution wraps the entire stem of the inner shell and fills the mold, and then put it in a room temperature dark place until it hardens;

[0080] (3) Cutting: Cut the embedding and hardening resin into 2-3mm resin slices along the cross section of the inner shell, and make the grinding surface parallel to the cutting surface of the inner shell;

[0081] (4) Fixing: stick the resin sheet (with the proximal end of the inner shell facing down) ...

Embodiment 3

[0090] Embodiment 3: A kind of method for observing and counting the growth pattern of the inner shell of soft fish

[0091] The operation steps are as follows:

[0092] (1) Picture production: Place the prepared section on a glass slide, and drop a drop of water with a rubber dropper (to increase the clarity of observation); under 200× magnification of an Olympus microscope connected to a CCD (CCD image sensor) Take pictures of the growth pattern of the inner shell; use Photoshop 7.0 image processing software to process the pictures taken respectively;

[0093] (2) Counting of growth lines: count the number of growth lines from the dorsal core of the inner shell to the ventral edge of the image collected at 200× magnification ( figure 2 ), during the counting process, select a place with a wider wheel spacing for counting ( figure 2 ).

Abstract

The invention specifically relates to a method for determining the age of Ommastrephidae by using an inner shell, belonging to the field of fisheries science. The method comprises grinding of the cross section of the inner shell and observation and counting of growth striations and comprises the following main steps: extraction, cleaning, dehydration, drying, tailoring, resin embedding, cutting, fixation, polishing, burnishing, picture making, counting of growth striations, etc. The method provided by the invention overcomes the problems that the inner shell is difficult to grind, grinding easily leads to shedding and growth striations are hard to observe and count; and the method realizes rapid and accurate grinding to an optimal plane, guarantees high success rate of slice preparation and can accurately determine the number of growth striations.

Description

technical field [0001] The invention relates to the field of fisheries, and relates to a method for observing the growth pattern of the inner shell, in particular to a method for identifying the age of the soft fish by using the inner shell. Background technique [0002] Squids are the general term for species of Molluscs, Cephalopoda, Lanceles, and Squids. Their lifespan is about 1 year old. They are important marine economic species and are widely distributed in oceans, Antarctica, and the North Pole. As a key content in fishery biology and ecology of soft fishes, age identification has been widely concerned by scholars at home and abroad. Otoliths, horny jaws, etc. are considered to be reliable materials for identifying the age of soft fishes. In the past, some studies tried to use the growth lines on the surface of the inner shell to identify the age of soft fishes, but it was difficult to interpret the early growth lines. It cannot be used for age identification. Con...
