[0030] Materials and methods

[0031] Mice: All experiments were performed according to the guidelines of the Experimental Animal Center of Sun Yat-sen University and approved by the Animal Ethics Committee. BALB / c was purchased from the Experimental Animal Center of Sun Yat-sen University (Guangzhou, China). PD-1H KO mice have been described in existing literature and were backcrossed with BALB / c mice to generate H-2 d / PD-1H KO strain. All experiments used 8-10 week old mice and were raised in SPF environment.

[0032] Production of PD-1H mAb: The full-length mouse PD-1H-pcDNA3.1 was stably transfected into Chinese hamster ovary cells (CHO cells) by lipofection, and the stable transfection was confirmed by flow cytometry using mam82mAb PD-1H + CHO cells. PD-1H-Ig fusion protein was generated and purified as described. PD-1H KO mice were immunized with mouse PD-1H-Ig, and hybridomas secreting PD-1H mAb were generated as described previously. Using PD-1H + The specif...

Abstract

The invention discloses application of PD-1H agonists to preparing medicine compositions for preventing or treating asthma of objects. The application has the advantages that treatment is carried out by the aid of agonist type PD-1H monoclonal antibodies, accordingly, the quantities of inflammatory cells (eosinophil and lymphocyte in particular) in mouse BALF (bronchoalveolar lavage fluid) are obviously reduced, pulmonary inflammation and myxopoiesis are obviously reduced, and accordingly the fact that PD-1H can be targeted for treating allergic diseases is indicated by results.

Description

technical field [0001] The present invention relates to a method of preventing or treating asthma in a subject. The present invention also relates to the use of the PD-1H agonist in the preparation of a pharmaceutical composition for preventing or treating asthma in a subject. Background technique [0002] Asthma is a common chronic inflammatory disease of the airways, studies have shown that CD4 + Th2 cells play a crucial role in the induction, mechanism and progression of diseases. The hallmark of a Th2-type response in asthma is eosinophilic airway inflammation with mucus secretion, airway remodeling, and hyperresponsiveness. IL-5 and IL-13 are important for the pathophysiology of asthma. IL-5 has multiple effects, including direct activation of eosinophils, effects on adhesion, and induction of synthesis of chemokines and regulators of inflammation. IL-13 primarily affects bronchial hyperresponsiveness, inflammation, and airway remodeling. Furthermore, IL-13 drives ...
