Embodiment Construction

[0040] The present invention will be described in detail below in conjunction with the accompanying drawings and specific embodiments.

[0041] A method for detecting AFP based on graphene, thionine and nucleic acid aptamer, the detection principle see figure 1 ,

[0042] The implementation steps are as follows:

[0043] 1. Electrode pretreatment: before using the screen printing electrode, put it in 0.5mol / LH 2 SO 4 In the solution, use the cyclic voltammetry scanning method to scan, the scanning voltage is -0.2 V ~ 1.0 V, and scan 10 times within this range; after the scanning is completed, the electrode is washed with distilled water and dried for later use;

[0044]2. Electrode modification and aptamer immobilization: Put the activated screen-printed electrode into 5mL0.01%HAuCl 4 In the solution, the electrodeposition was carried out while stirring with a magnetic stirrer, the deposition time was 120 s, and the deposition potential was -0.5 V. After the deposition was...

Abstract

The invention discloses a method used for detecting alpha fetoprotein based on graphene, thionine, and a nucleic acid aptamer. According to the method, electrodeposition technology is adopted for deposition of nano-gold on the surface of a screen-printed electrode, electrostatic adsorption is adopted for absorption of graphene and thionine which drop on an electrode surface onto the surface of thescreen-printed electrode, thionine is taken as a bridge molecule for capturing of the nucleic acid aptamer onto the surface of a modified electrode. A nanometer aptamer biosensor can be constructed based on the extremely large specific surface area and signal amplification effect of graphene, the high loading capacity of thionine on the aptamer, and the specific recognition effect of the aptameron alpha fetoprotein. Peak value current is measured through differential pulse voltammetry, and the relation curve of the current to the alpha fetoprotein concentration is drawn, so that detection onalpha fetoprotein is short in time, low in cost, and high in the specificity.

Description

technical field [0001] The invention belongs to the technical field of biological detection, and in particular relates to a method for detecting alpha-fetoprotein. Background technique [0002] Alpha-fetoprotein (Alpha-fetoprotein, AFP) is a glycoprotein of the albumin family, which is a single polypeptide chain formed by more than 590 amino acid residues. The content of alpha-fetoprotein in normal adults is very small, but in liver cancer The content of alpha-fetoprotein in the patient's body is related to the degree of differentiation of hepatocellular carcinoma. AFP detection methods mainly include enzyme-linked immunosorbent assay, fluorescent immunoassay, electrochemiluminescence and electrochemical immunosensor. However, conventional immunoassays have their inherent disadvantages, such as radioactive contamination in enzyme-linked immunoassays, the activities of enzymes and antibodies used in immunoassays are easily affected by external temperature and pH, unstable on...
