Problems solved by technology

Chronic pancreatitis is misdiagnosed as pancreatic cancer during pancreatic cancer detection, which will lead to further overdiagnosis and even overtreatment, bringing unnecessary physical, psychological and economic burdens to patients
However, in the existing studies, they often focus on the sensitivity of pancreatic cancer detection, ignoring the problem of high false positives caused by misjudgment of pancreatitis that may exist in the detection, which brings unnecessary physiological harm to patients. , psychological and economic burden

Embodiment 1

[0069] Plasma samples from 19 patients with pancreatic cancer, 15 patients with chronic pancreatitis, and 16 normal subjects were obtained. All samples come from Boercheng Company. Pancreatic cancer, chronic pancreatitis, and normal human genomic DNA were then extracted, and the genomic DNA samples were pretreated so that unmethylated cytosine bases at the 5' position were converted to uracil, thymine, or Another base not used for cytosine in hybridization behavior. In this example, this pretreatment was achieved by bisulfite reagent treatment. The extraction and processing of the DNA can be carried out by any standard means in the prior art. Specifically, in this embodiment, all sample DNA extraction and bisulfite DNA modification are carried out by using Boercheng Company Extracted from the plasma processing kit. Then, the above-mentioned BNC1 and ADAMTS1 gene primers, probe combinations and internal reference gene ACTB gene primers and probe combinations were added to th...

Abstract

The present invention provides a composition for in vitro detection of pancreatic cancer / chronic pancreatitis, wherein the composition comprises: 1) a nucleic acid for detecting the methylation levelin at least one region of a target gene, wherein the target gene is selected from a basonuclin 1 gene or a fragment thereof, and a human a-disintegrin-and-metalloproteinase-with-thrombospondin-motifs1 gene or a fragment thereof, 2) a nucleic acid for detecting at least one region of beta-actin gene, and 3) a reagent for detecting the concentration of a CA-199 antigen in serum / plasma. The invention further provides a kit comprising the composition, and a method for in vitro detection of pancreatic cancer. With the method of the present invention, pancreatic cancer and non-pancreatic cancer (including normal condition and chronic pancreatic cancer) can be effectively distinguished, such that the misjudgment of chronic pancreatitis is avoided, the accuracy of the detection is improved, and the clinical applicability is effectively enhanced; and the non-invasive and rapid in vitro detection method for pancreatic cancer is provided.

Description

technical field [0001] The invention belongs to the field of medical detection and relates to the use of a composition in disease detection, in particular to the use of a composition and its corresponding kit in preparing a device for distinguishing pancreatic cancer and chronic pancreatitis. Background technique [0002] Pancreatic cancer is a highly malignant tumor of the digestive tract that is difficult to detect and treat. The reason for the high mortality rate of pancreatic cancer is that the diagnosis rate of early pancreatic cancer is extremely low. The process from the formation of pancreatic cancer lesions to the appearance of clinical symptoms takes an average of 2-3 years. Making full use of this window period to improve the diagnosis rate of early pancreatic cancer is expected to improve the treatment effect of pancreatic cancer and reduce the mortality rate of pancreatic cancer. The Chinese patent application "composition for detecting pancreatic cancer and i...
