Abstract

The invention relates to a preparation method and application of an algae solution of euglena gracilis klebs for determining the biotoxicity of particulates in water, and belongs to the field of environmental health toxicology. The preparation method comprises the following steps of culture of algae species and preparation of the algae solution, and specifically comprises the following steps: selecting the algae species of the euglena gracilis klebs, inoculating a container which contains a liquid culture medium with the algae species under a sterile condition, completing inoculating, culturing in a constant-temperature illumination incubator until a logarithmic phase, and standing for standby use; carrying out dark treatment on the algae solution which is cultured to the logarithmic phase, and determining OD680nm value by using a spectrophotometer or eliasa; drawing a cell growth curve according to the OD680nm value determined in step B, and selecting the algae solution of the euglenagracilis klebs of which the cell density range is 1*10<5> to 1*10<6> in the logarithmic phase per milliliter, thus determining the biotoxicity of the particulates in the water. The invention also provides determination of the algae solution of the euglena gracilis klebs in determining the biotoxicity of the particulates in the water. Through the preparation method provided by the invention, the biotoxicity of the particulates in the water can be directly expressed.
