Problems solved by technology

However, plants are affected by a variety of environmental factors under natural conditions, and the morphological configuration of the root system is relatively complex. Therefore, the data obtained by soil in-situ observation and measurement cannot accurately reflect the influence of a single environmental factor on root growth (Ma Xinming et al., 2003; Zhu et al., 2006)

Embodiment 1

[0035] Example 1 In situ observation and determination of the morphological configuration of the Arabidopsis root system under aluminum stress

[0036] 1. Design transparent cultivation containers and growth media;

[0037] as attached figure 1 As shown, use a sterile square petri dish with an aspect ratio of 10cm×10cm and a height of 1.5cm;

[0038] The transparent medium is an improved solid medium, which is a mixture of plant growth nutrient solution and 0.8% agar (sigma; A1296), and the pH value is adjusted to 5.8 according to the conventional techniques of those skilled in the art.

[0039] The processing medium is ordinary qualitative filter paper; aluminum processing includes aluminum-free control or 50uM ACl 3 For the treatment, refer to the conventional technique of those skilled in the art, and adjust the pH value to 4.2; according to the conventional technique, reagents such as hydrochloric acid or sodium hydroxide are used for adjustment.

Abstract

The invention discloses a sterile filter paper cultivation method for studying interaction between plant roots and environment. The method comprises following steps: S1. designing visual cultivation system, including a transparent cultivation container, a treatment medium, a triangular support and a growing medium; S2. sterilizing plant seeds and germinated, and after germination, transplanting the seedlings into a cultivation container to keep the plant growing naturally in the growth medium; S3. carrying out biotic or abiotic stress treatment on the germinated plant seeds, and tilting culture; S4. scanning roots by a camera system, a stereomicroscope and a scanner to perform two-dimensional in situ observation and image capture of roots. This method can directly perform 2-D visualization on the in-situ growth of plant roots without changing the growth environment of plant roots and its surroundings; combined with computer image analysis, in-situ observation and measurement of root response to the environment are carried out, and dynamic, in-situ and non-destructive root-environment interaction is observed and measured.

Description

technical field [0001] The invention belongs to the technical field of plant physiology. More specifically, it relates to a sterile filter paper cultivation method for studying plant root-environment interactions. Background technique [0002] The root system is the main organ for plants to absorb nutrients and water, and also the main organ for material and signal communication between plants and the soil environment (Barber, 1995; Bailey et al, 2002; Lynch, 2007; White et al., 2010; Vacheron et al. , 2013; Hasan et al., 2016; Hippler et al., 2018). Therefore, the study of plant root growth is of great significance to understand the nutrient requirements of plants and their interactions with the environment. [0003] Under natural conditions, due to the opacity of the soil medium, in situ observation of the root system requires large-scale excavation of the plant root system from the soil. However, plants are affected by a variety of environmental factors under natural c...
