Abstract

The iris ensata root induced callus culture method includes disinfecting mature seeds; inoculating the mature seeds in seed germination culture media with 0.5 mg / L of MS+6-BA and the seed germination rates of approximately 100%; inoculating each seedling into rooting culture media when the seedling with 2-3 leaves grows, and allowing the seedlings to grow to obtain aseptic seedlings with developed root systems; cutting root segments of maturation zones with fibrous roots (partially cutting off the fibrous roots and exposing wound) into induction culture media. The lengths of the root segments inoculated into the inductionculture media are 1.0-1.5 cm. The optimal induction culture media are 1.0 mg / L of MS+6-BA, 2 mg / L of 4-D and 0.2 mg / L of NAA, and the induction rate of the induction culture media can reach 100% on the 30th day.
