Problems solved by technology

Antibiotics are currently a common method for the treatment of Aeromonas hydrophila infection, but due to the large and unreasonable use of antibiotics, the emergence of multidrug-resistant Aeromonas hydrophila is becoming more and more frequent, which not only easily leads to the failure of antibiotic treatment Failure, more likely to lead to food safety issues with fish products

[0076] 1. Isolation and purification of phage

[0077] Sample collection and processing: Collect water bodies in fish ponds with Aeromonas hydrophila disease or domestic sewage, the volume is 500mL, refrigerated transportation, take 200mL after aseptic filtration in the laboratory, and mix with Aeromonas hydrophila 2016- 76 were inoculated together in TSB liquid medium and cultured with shaking at 18°C for 2 days to obtain the phage proliferation solution. Distilled water was used as a control. The results showed that the culture medium in the domestic sewage group became clear, while the bacteria in the distilled water control group grew normally ( figure 1 Middle A)

Abstract

The invention discloses bacteriophage pAhMJG and application of the bacteriophage pAhMJG in treatment of fish diseases caused by aeromonas hydrophila. The preservation number of the bacteriophage pAhMJG preserved in the China Center for Type Culture Collection is CCTCC No: M 2019202. The whole length of a bacteriophage pAhMJG genome is 45,057 bp; and the genome is a linear double-stranded DNA andcontains 45 open reading frames. The bacteriophage pAhMJG does not contain sRNA, tRNA or rRNA, also does not contain antibiotic genes, virulence genes or lysogen related genes, has an obvious killingeffect on aeromonas hydrophila, can be used for treating fish diseases caused by aeromonas hydrophila, and has a wide application prospect.

Description

technical field [0001] The invention relates to the field of biotechnology, bacteriophage pAhMJG and its application in treating fish diseases caused by Aeromonas hydrophila. Background technique [0002] Aeromonas hydrophila (Aeromonas hydrophila, Ah) belongs to the Aeromonas family (Aermonadaceae) Aeromonas (Aeromonas). The bacterium is a Gram-negative brevibacterium, which widely exists in various water bodies, soils and aquatic products in nature, and can cause systemic sepsis in molluscs, fish, amphibians, reptiles and mammals, etc. Or local infection, often lead to a large number of animal deaths, but also bring serious economic losses to the aquaculture industry. Antibiotics are currently a common method for the treatment of Aeromonas hydrophila infection, but due to the large and unreasonable use of antibiotics, the emergence of multidrug-resistant Aeromonas hydrophila is becoming more and more frequent, which not only easily leads to the failure of antibiotic treat...
