The method comprises the following steps: taking natural abundance carbon dioxide as a raw material, and carrying out a first gas diffusion cascade; obtaining carbon dioxide of which the carbon-13 isotope abundance is higher than 45% at the heavy fraction end of the first gas diffusion cascade, supplying the carbon dioxide serving as a supply material of a second gas diffusion cascade into second gas diffusion cascade, and obtaining a carbon dioxide product of which the carbon-13 isotope abundance is higher than 90% at the light fraction end of the second gas diffusion cascade; wherein the first gas diffusion cascade and the second gas diffusion cascade are both in relative abundance matching cascade. The method is realized based on gas diffusion cascading, has the characteristic of large flow, and is convenient for chemical amplification.
