Problems solved by technology

[0004] Fish odor syndrome is caused by a missense mutation of the flavin monooxygenase 3 (FMO3) gene located on chicken chromosome 8, which causes the body to fail to decompose trimethylamine in food normally. The mutation is located at the seventh position of the FMO3 gene. On the exon, the 1034th base adenine nucleotide (A) of the mutant individual cDNA sequence is replaced by thymine (T), which makes the 329th amino acid of the encoded protein changed from threonine (T) to serine (S), leading to loss of enzyme function

Embodiment 1

[0025] Example 1 Cultivation and acquisition of pure line of green-shell layer hens without fishy smell

[0026] Refer to the aforementioned method of molecular marker-assisted cultivation of green-shell layer hens with green feet and hemp feathers without fishy smell, the details are as follows:

[0027] From 7,500 heterozygous hens of Xinyang green-shell hens (black feather, white skin, green feet, green-shell eggs) introduced from Shanghai Xinyang Poultry Breeding Center, 1,000 individual hens with good appearance and body shape were selected. Crossed with 80 roosters of Yao chicken strain (black hemp feather, white skin, green feet, pink shell eggs) owned by Guangxi Guigang Gangfeng Agriculture and Animal Husbandry Co., Ltd. in a mixed way, collected breeding eggs, hatched, and obtained 10040 F1 Generation chicks.

[0028]During the breeding process, the unqualified, deformed and unhealthy individuals were eliminated at any time. When the F1 generation was raised to 4 wee...

Embodiment 2

[0035] The fishy smell and the purity test of the green-shell egg performance of the pure line of green-shell hens without fishy smell obtained in Example 2

[0036] The pure line of green-shell layer hens without fishy smell obtained in Example 1 was cross-crossed, and 2000 first chicks of pure line F3 offspring were obtained. Blood was collected from all individuals at the age of 4 weeks, and after DNA extraction, the genotype purity of fishy smell and green-shell egg traits was identified at the molecular level; the eggshell color of eggs laid by pure line offspring hens was identified after breeding until the peak egg production period.

[0037] It was found (Table 2) that at the molecular level, the genotypes identified at the age of 4 weeks without fishy smell and green shell egg traits were all homozygous ( figure 2 A and 2B); When feeding to the peak egg production period, among the 891 individual hens that laid eggs, the eggshell colors of the eggs produced were all ...

Abstract

The invention discloses a method for culturing green-foot pock-feather green-egg-shell laying hens through molecular marker assistance. A Yao chicken strain and a green-egg-shell laying hen strain arehybridized, a strategy of gene pyramiding is adopted, molecular markers of related genes of fish smell and green-egg-shell properties are combined to implement screening, and finally properties of green shell eggs without fish smell are achieved while a pure strain is obtained. Experiments show that by adopting the method, rapid breeding of novel chicken strains with multiple excellent properties(no fish smell and green shell eggs) can be successfully achieved. The method can be applied to rapid and simple culture on a matching strain of no-fish-smell green-egg-shell laying hens with green shell egg properties without fish smell, and property research and production of related matching strains; and in addition, a combined identification method of molecular markers is provided for fish smell and green shell egg properties, and the purity of a no-fish-smell green-egg-shell laying hen strain can be verified and analyzed. Compared with the prior art, the method has the characteristics ofbeing rapid, accurate, convenient and rapid, efficient, economic, and the like.

Description

technical field [0001] The invention belongs to the technical field of poultry breeding, and in particular relates to a method for molecular marker-assisted cultivation of green-shell laying hens with green legs and hemp feathers without fishy smell. Background technique [0002] In recent years, nutritionists and medical scientists at home and abroad have concluded that the nutritional value and health care functions of eggs mainly include five aspects: strengthening the brain and improving intelligence, protecting the liver, preventing arteriosclerosis, preventing cancer, and delaying aging. On the basis of the above functions, green-eggs play an extremely important role in repairing human tissues, forming new tissues, consuming energy and participating in complex metabolic processes because they contain a large amount of essential vitamins and minerals and high biological value proteins. important role. With the improvement of the production performance of laying hens an...
