Embodiment

[0033] A method for predicting coagulation abnormalities during pregnancy in a population with recurrent miscarriage, comprising the following:

[0034] 1. Research and experimental plan

[0035] A total of 141 patients with URSA were selected as the research subjects, and their pregnancy, pregnancy outcomes and neonatal conditions were followed up. Blood samples were collected and questionnaires were used to collect past medical history, family medical history, reproductive history, socioeconomic status, occupational / environmental exposure, psychological stress, anxiety, smoking / passive smoking / drinking, drug use, and lifestyle. (including exercise), URSA-related drug use (glucocorticoids, folic acid, progesterone, aspirin, and low molecular weight heparin, etc.), sleep status, and physical examination / pregnancy test data.

[0036] Platelets from plasma were isolated by differential centrifugation, then the platelet pellet was digested with nuclear genomic DNA interference u...

experiment example

[0050] 1 Basic characteristics of the study population

[0051] The baseline characteristics of the selected 141 URSA subjects are shown in Table 1. The average levels of D-Dimer, PS, PC, and HCY were 350.2±438.4, 74.04±18.95, 105.1±18.03, and 9.511±6.682, respectively, all of which were in the normal diagnosis. Within the range, no abnormal coagulation during pregnancy occurred.

[0052]

[0053] 2 The relationship between a single platelet mtDNA locus and abnormal coagulation function during pregnancy

[0054] In the follow-up population, the single platelet mtDNA loci associated with abnormal coagulation function are shown in Table 2, involving a total of 18 loci (ATP8p1, ATP8p2, ATP8, COX1p1, COX1p2, COX1, COX2p1, COX2p2, COX2, COX3p1, COX3p2, COX3, ND5p1, ND5p2, ND5, TRNAp1, TRNAp2, TRNA). The results showed that there was no statistically significant difference between abnormal coagulation during pregnancy and the above single platelet mtDNA locus (p>0.05).

Abstract

The invention discloses a method for predicting abnormal blood coagulation function of recurrent abortion crowds during pregnancy, which comprises the following steps: separating platelets in plasma of URSA patients by using a differential centrifugation method, then carrying out nuclear genome DNA interference treatment digestion on platelet precipitation by using DNA restriction enzyme, then directly carrying out bisulfite treatment on digestion products, and finally predicting the abnormal blood coagulation function of the recurrent abortion crowds during pregnancy. A pyrosequencing method is combined to detect the methylation level of respiratory chain related gene loci, including MT-ATP8, MT-COX1, MT-COX2, MT-COX3, MT-ND5 and MT-TRNA, in the platelet mtDNA; the method has the advantages that the methylation level is detected by selecting the mitochondrial sites related to URSA, and the diagnosis efficiency of the biomarker is evaluated by using AUC, so that the prediction capability on the onset risk of the blood coagulation dysfunction is improved.

Description

technical field [0001] The invention relates to a method for predicting the abnormal coagulation function of the recurrent miscarriage population during pregnancy, in particular to a method for predicting the abnormal blood coagulation function of the recurrent miscarriage population during pregnancy by using epigenetic markers, and belongs to the field of genetics. Background technique [0002] Spontaneous abortion accounts for 15%-40% of all pregnancies, and 2 or more consecutive spontaneous abortions are called recurrent spontaneous abortion (RSA). The factors that cause RSA are complex and diverse, including genetic factors, infectious factors, Endocrine factors, anatomical factors, immune factors and coagulation factors. Among them, 40% to 50% of patients have unexplained miscarriage, which is called unexplained recurrent miscarriage (URSA). Abnormal coagulation function is considered to be the main cause of RSA. Studies have shown that nearly 55%-62% of patients with...
