Embodiment

[0019] 1. Sampling mechanical method Harvest bloom cyanobacteria from eutrophic water body, obtain fresh cyanobacteria algae paste (conventional method) after concentration and dehydration, and the water content of algae paste is about 92%.

[0020] 2. Extract the crude extract of cyanobacteria protein: take fresh cyanobacteria, after dehydration, add 0.05M-0.2M phosphate buffer solution (pH 5.0 -9.0), mix well, and then freeze at -15°C. After freezing, take it out and place it at room temperature at 20°C or 22°C or 25°C to thaw it. The crude extract is extracted from the cell, and then the crude extract is separated from the cell fragments by plate-and-frame filtration or centrifugation to obtain the crude extract of cyanobacteria protein.

[0021] 3. Microfiltration and pretreatment: the crude extract of cyanobacteria protein in the above step is passed through two kinds of microfilters with pore diameters of 2-3 μm and 0.45-0.22 μm in sequence, and then undergoes microfiltr...

Abstract

The invention discloses a method for preparing phycocyanin by using bloom cyanophyte. Said invention uses the fresh bloom cyanophyte as raw material, and adopts the following steps: first step, extracting crude filtrate; dehydrating fresh bloom cyanophyte, adding buffer solutio, mixing them uniformly, frezing, takingout and thawing at room temperature, freezing and thawing for 3-5 times and filtering by using plate and frame; second step, ultrafiltering and purification; using hollow fibre ultrafiltration device whose trapping molecular weight is 1000-200000 daltons to remove various substances with smaller molecular weight from crude extract passed through microfiltration by using ultrafiltering membrane; third step, settling-clearing purification; removing most of small molecules from phycocyanin portion purified by means of ultrafiltration, at the same time making concentration and storing the concentrated phycocyanin portion at a certain temperature; and fourth step; hydroxyapatite column chromatography to make purification.

Description

technical field [0001] The invention relates to extracting natural blue pigment from cyanobacteria in water bloom, more specifically to a method for preparing phycocyanin from cyanocyanin in water bloom, which is suitable for coloring (coloring) coloring agent of food, beverage and cosmetics. Background technique [0002] In recent years, due to the rapid development of industry, the rapid increase of population, the aggravation of urbanization, and the severe environmental pollution, the degree of eutrophication in many water bodies in my country has intensified, causing a large number of algae, especially blue-green algae, and some water bodies even have serious blue-green algal blooms. Hua, directly affects the quality and availability of water bodies, and endangers the health of livestock and human beings. A large number of cyanobacteria blooms in lakes in the middle and lower reaches of the Yangtze River occurred from April to October, especially i...
