Abstract

The invention relates to a concentration and separation method of microcomponent in a complex system, in particular to the selective concentration and separation method of genistein in radix puerariae crude extract. The steps adopted are as follows: a) genistein molecularly imprinted polymer microspheres are employed as filler to prepare solid phase extraction column; b) the solid phase extraction column prepared in the step a is activated and sampled and then the solid phase extraction column is leached by using acetonitrile solution with water accounting for 49-51 percent of volume ratio and acetonitrile solution with water accounting for 4.9-5.1 percent of volume ratio in sequence, and then the leached solid phase extraction column is eluted by methanol solution with water accounting for 0.18-0.22 percent of volume ratio, thus obtaining finished products through drying the eluent. The invention has the advantages that: after the radix puerariae ethanol crude extract is extracted through molecularly imprinted solid phase, a clean chromatogram is obtained after removing most disturbance factor and the recovery ratio of the genistein reaches above 70 percent. Meanwhile, glycitein is reserved obviously. The invention has wide application prospect on the pretreatment of radix puerariae crude extract samples.
