The method is characterized by applying oral oxytocic hormone to rice field eels, namely feeding parent rice field eels with a certain dosage of oxytocic hormone by using animal bait as a carrier of the oxytocic hormone; and the method comprises the following steps of: stopping feeding of both male and female parent rice field eels for 1 day before spawning induction; observing oestrus conditions of bubbling and nesting of the male and female rice field eels in 3 to 4 days after the male and female rice field eels ingest an oxytocic hormone liquid medicament, and surrounding and marking nests with eichhornia crassipes after the nesting of the parent rice field eels is finished, wherein an inner net cage in a paddy field is used as a spawning pond; and naturally inseminating the parent rice field eels in the spawning pond, breaking membranes by fertilized ova and emerging seedlings after 144 hours, inhabiting all fish fries in the eichhornia crassipes surrounding the nests of the parent rice field eels after the fish fries break the membranes after 96 hours, and collecting the rice field eel seedlings at the time.
