Embodiment Construction

[0029] The invention is used to detect leukemia susceptibility gene combination, which includes the combination of three genes closely related to leukemia: CYP1A1 gene and NQO1 gene of poison metabolism and XRCC1 gene of DNA repair. Including the following SNP sites: rs4646903 site of CYP1A1 gene, rs1800566 site of NQO1, rs1799782 and rs25487 site of XRCC1.

[0030] The gene combination for detecting leukemia susceptibility is used for designing specific primer pairs and probes for the site.

[0031]The primers described in the present invention are designed for the rs4646903 site of the CYP1A1 gene, the rs1800566 site of NQO1, and the rs1799782 and rs25487 sites of XRCC1, and can be used for multiplex PCR amplification, and can simultaneously amplify the four sites above Gene fragment. Designing such primers is readily accomplished by those skilled in the art. Preferably, the primers described in the present invention have the following sequence:

Abstract

The invention discloses a gene combination, a primer and a probe for detecting leukemia susceptibility and application. The gene combination comprises a combination of three genes closely related with leukemia, namely a CYP1A1 gene and an NQO1 gene of poison metabolism and an XRCC1 gene of DNA repair. The gene combination comprises the following SNP sites: rs464603 site of the CYP1A1 gene, rs1800566 site of the NQO1 gene, and rs1799782 and rs25487 sites of the XRCC1 gene. Whether the detected crowds carry 'leukemia susceptible genes' are comprehensively detected and analyzed by detecting a group of genes and sites related with the leukemia susceptibility, using the specific primer and the probe and combining a mononucleotide extending technique and a micro array chip technique so as to screen the leukemia susceptible crowd from the crowds, change unhealthy lifestyles and fulfill the purpose of preventing.

Description

technical field [0001] The present invention relates to a gene combination, primer, probe and use, especially a gene combination, primer, probe and use for detecting leukemia susceptibility. Background technique [0002] Leukemia has a high incidence rate in the Chinese population and is the most common malignant tumor in children and young adults. In recent years, its incidence rate is on the rise, and the etiology has not yet been elucidated. The current accepted view is that leukemia is caused by environment-gene interaction. Genetic aspects involve mutations in metabolism, DNA damage repair genes, etc. Studies have found that the metabolic enzyme family in the carcinogen metabolic pathway is polymorphic in the population, and different individuals have huge differences in their ability to metabolize specific carcinogens. Likewise, there are significant individual differences in DNA repair capacity. Human Genome Project research results show that everyone's genes are ...
