Abstract

The invention discloses a sterile dynamic detecting method for tissue culture seedling nitrogen use ratio, which comprises the following steps of: constructing a water loss model and calculating the thickness of culture medium; collecting the diffuse reflection spectrum information in multi-point manner of the culture medium in the modeling sample and the test sample by the near infrared spectrometer to acquire the model of the modeling sample; then forecasting the test sample and acquiring the multipoint nitrogen content in the culture medium of the test sample; carrying out linear fitting by the nitrogen content of a certain point and the vertical distance between the point and the base diameter of the tissue culture seedling to acquire an equation between the nitrogen content and the vertical distance; calculating the total nitrogen amount of the culture medium of the test sample according to cylinder integral formula; detecting the multipoint nitrogen content in the culture mediumof the test sample and detecting the biomass of the tissue culture seedling in the test sample by image processing method at the same time; and finally calculating the nitrogen use ratio of the tissue culture seedling in the test sample according to the formula. The change of nitrogen in the culture medium in the culturing process of plant tissue can be measured in dynamic, sterile and undamaged way without extracting the nitrogen from the culture medium by the sterile dynamic detecting method.
