Embodiment Construction

[0019] The present invention will be described in detail below in conjunction with embodiments.

[0020] 1. Sema4D calmodulin-binding peptide inhibits the proliferation of HeLa cells

[0021] In this experiment, human cervical cancer HeLa cells were selected, and the effect of Sema4D calmodulin-binding peptide on the proliferation of HeLa cells in vitro was preliminarily explored. HeLa cells were inoculated in RPMI-1640 medium containing 10% fetal bovine serum, 100 U / ml penicillin, and 100 μg / ml streptomycin, and cultured in a 37 °C, 5% CO2 incubator. The medium was changed every 48 hours and subcultured once, and the cells in the logarithmic growth phase were selected for the experiment.

[0022] step:

[0023] 1) Cell inoculation: Digest the cells in the logarithmic growth phase with 0.25% trypsin, prepare the cells into a single-cell suspension with complete medium, and inoculate them in a 96-well plate at an appropriate density, 100ul per well;

[0024] 2) Cultivate cel...

Abstract

The invention discloses a CaM binding peptide, which belongs to an amino acid and is in the molecular type of peptide. The CaM binding peptide is a Sema4D calmodulin binding peptide synthesized in a laboratory, and contains a sequence of which the code corresponds to Sema4D binding CaM. The molecular structural formula of the Sema4D calmodulin binding peptide is NH2-GYLPRQCLKFRSALLIGKKKPKS-COOH. The CaM binding peptide is applied to tumor resistance and inhibition of in-vitro angiogenesis of an HUVEC (Human Umbilical Vein Endothelial Cell). The CaM binding peptide can possibly become a novel angiogenesis inhibitor, and a novel antitumor medicament is provided for clinical use.

Description

[0001] technical field [0002] The invention relates to the field of anti-tumor polypeptide drugs, in particular to a CaM-binding peptide and its application. Background technique [0003] At present, the rampant cancer is threatening people's health. Overcoming persistent diseases is a major problem facing mankind. Today, with advanced science, people still focus on finding drugs with strong anti-cancer effects and few side effects to deal with tumors. A large number of data confirm that intracellular calmodulin (CaM) is closely related to the formation, growth and proliferation of tumors. [0004] In the 1970s and 1980s, it was confirmed that calmodulin antagonists have certain inhibitory effects on tumor proliferation and metastasis. CaM acts as intracellular Ca 2+ Receptor protein from, involved in intracellular Ca 2+ Calcium is required for normal cell proliferation, so it is believed that CaM is necessary for normal cell proliferation. It has been found that the...
