Abstract

The invention relates to plant propagation, namely a one-step rapid seedling method for a tender stem segment of malus komarovii inside a test tube. The method comprises the steps of (1) collecting branches of the malus komarovii in the last ten days of October, carrying out dormancy at low temperature; promoting a dormant bud to germinate by water culture, and cutting the processed branches into segments, each of which contains a leaf, as explants for use; (2) enabling the malus komarovii to root, wherein the culture medium for growth of an axillary bud of the stem segment is culture medium+6-BA (0.07-0.09mg.L-1)+IBA (0.02-0.04mg.L-1)+GA3(0.09-0.13mg.L-1). By adopting the one-step rapid seeding method disclosed by the invention, rooting of the tender stem segment of the malus komarovii inside the test tube is facilitated, and germination of the axillary bud is facilitated at the same time; the method is high in rooting percentage, rapid in germination speed of the axillary bud, and strong in maneuverability; just one low-cost culture medium is utilized from inoculation of the stem segment to germination; and the one-step rapid seeding method belongs to a one-step seeding method, and can meet the requirement of industrialized seedling production.
