Problems solved by technology

However, these works are all in their infancy, and no comparatively systematic research results have been formed, and there are still problems of severe browning of the induced callus and low regeneration rate (Li Ruifen, Sun Zhenyuan, Wei Jianhua, etc. Bison grass young ears Callus induction and plant regeneration[J]. Wuhan Botanical Research, 2004(5): 449-454.)

Embodiment 1

[0027] Example 1 Optimization of 2,4-D concentration in callus induction medium

[0028] 1. Pretreatment of seeds: taking out seeds from glumes;

[0029] 2. Disinfection of explants: first put the washed seeds into a sterile container, sterilize with 70% alcohol for 60 seconds in a sterile workbench, then sterilize with 7-10% sodium hypochlorite solution for 30 minutes, and use sterile Rinse with water 4-5 times before use.

[0030] 3. Germination of seeds: first configure MS medium without hormones, sterilize it and pour it into a large petri dish to cool for later use, and place the sterilized seeds in a large petri dish with hormone-free MS medium. Afterwards, the petri dish was sealed and placed in a light incubator for dark cultivation for 2-3 days.

[0031] 4. Induction of mature embryo callus: select seeds that have germinated and grown radicles from the petri dish, and transfer them to the callus induction medium for callus induction (see figure 1 , figure 2 ). T...

[0034] Example 2 Optimization of Germination of Seeds

[0035] 1. Pretreatment of seeds: taking out seeds from glumes;

[0037]3. Germination of seeds: first spread double-layer filter paper in the petri dish, after autoclaving for 20 minutes, place it in a sterile operating table, moisten it with sterile water, place the sterilized seeds on the sterile filter paper, and Add some sterile water to the Petri dish to keep the seeds hydrated for germination. Afterwards, the petri dish was sealed and placed in a light incubator for dark cultivation.

Abstract

The invention provides an inducing method of a Buchloe dactyloides callus. The inducing method adopts a Buchloe dactyloides seed as an explant, the Buchloe dactyloides seed is disinfected to be arranged on a sterile filter paper for germination, selecting germinated seeds with radicle, and placing into a callus induction medium for culturing for 20-25 days at temperature of 25 + / -1 DEG C, wherein the callus induction medium is a solid medium with pH value of 5.8-6.0 and is prepared from the following components: MS medium, 3.0-3.5mg / L of 2,4-D (2,4-Dicholrophenoxyacetic acid), 30g / L of saccharose and 7g / L of agar. The method can eliminate the phenomenon that the seeds are not disinfected thoroughly during disinfection process and cause contamination during callus induction, also eliminates interference of dead seeds on the callus induction, reduces the operation difficulty, removes a step of vertically cutting a mature embryo, improves the callus induction rate up to more than 90% under a condition with simplicity and convenience in operation.

Description

technical field [0001] The invention belongs to the field of plant tissue culture, and in particular relates to a method for inducing callus of bison grass. Background technique [0002] Bison grass (Buchloe dactyloides) is a warm-season turfgrass with well-developed stolons. Bison grass has strong drought tolerance, heat resistance and flood tolerance, strong disease resistance and regeneration ability, and wide adaptability. It is a kind of warm-season turfgrass that is widely used at present. However, bison grass also has some disadvantages, such as the slow speed of seed building, the poor quality of the formed lawn, gray-green color, short green period in the north, poor shade tolerance and trampling resistance, and poor viewing and landscape effects at close range. Poor, dry and yellow in winter, etc., these shortcomings limit the application of bison grass in our country. In order to solve the problems in the application of bison grass, turf scientists have strength...
