Embodiment 1

[0138] Example 1 - Connectivity Mapping Approach to Identify Compounds with Retinoic Acid-Like Gene Regulatory Behavior

[0139] The connectivity mapping (Cmap) method was used to identify retinoid-like compounds.

[0140] Gene expression profiles were generated on skin biopsies obtained from healthy female subjects who were topically applied at a concentration of 0.025% ( ex. Johnson & Johnson) retinoic acid (all trans retinoic acid). Gene signatures derived from retinoic acid-treated skin were queried in the Cmap database. Identified compounds ranked high in Cmap were topically applied to skin grafts obtained from breast surgery. Gene arrays were repeated on samples treated 24 hours after topical application.

Embodiment 11

[0141] Example 1.1 Materials and methods

[0142] Treat your skin with retinoic acid

[0143] Skin biopsies were taken from the sun-exposed forearms of 8 healthy Causcasion female subjects aged 50+. Within 1 week, use retinoic acid (concentration of 0.025% ) to treat patients. At the end of 1 week, 3 mm punch biopsies were taken from each treatment site. The study was vehicle controlled. Two biopsies were taken per treatment in each arm to achieve sufficient RNA concentration. Biopsies were stored under controlled conditions at 4°C for 24 hours immediately prior to RNA extraction.

[0144] Treatment of skin grafts with retinoic acid

[0145] Subcutaneous fat was removed from full thickness skin (surgical waste from the breast reduction procedure) using a scalpel. Three Causcasian female donors aged 23, 41 and 60 were used in this study. These 3 female donors were different from those in the previously outlined skin treatment studies. 8 mm punch biopsies were ta...

Embodiment 12

[0153] Example 1.2 Results - Gene signature identification compounds using retinoic acid

[0154] Results of Cmap analysis link genetic signatures of retinoic acid-treated human skin to novel potential compounds with retinoic acid-mimicking activity

[0155] A number of genes were independently shown to be regulated by all trans retinoic acid. Due to differences between variables in patient material from graft studies and clinical studies, separate approaches were taken. Gene expression profiles in all trans-retinoic acid-treated biopsies compared to vehicle-treated controls were examined by DNA microarray analysis.

[0156] Two separate studies of gene expression profiles were performed in human skin topically applied all-trans retinoic acid. The first in vitro study was performed by topically applying all trans retinoic acid to skin grafts obtained from 3 donors within 24 hours, followed by microarrays. The second study was a clinical in vivo study in which all trans re...

Abstract

Non-therapeutic cosmetic or dermatological use of at least one of a compound selected from the group consisting of: gibberellic acids, naringenins, N-acetylaspartic acids, beta-aescin, arachadonic acid, quercetin, vitexin and docosahexaenoic acid (ethyl ester). Suitably, the use is in the treatment of, and / or prevention of, at least one sign of skin ageing or at least one sign of a skin damage condition associated with ageing, wherein the sign of skin ageing or skin damage is present on skin of the face, body or the scalp of a subject.

Description

technical field [0001] The present invention relates to compounds, or plant extracts comprising the same, wherein said compounds counteract the effects of skin aging and / or provide anti-aging benefits. More specifically, the present invention relates to a retinoic acid-like effect on the skin without the harsh effect and skin irritation caused by the use of retinoic acid. Background technique [0002] Aging is a multifactorial phenomenon. Skin aging is primarily the result of a person's genetic predisposition (also known as chronological aging) and a person's physiological response to environmental stress (also known as photoaging). Photoaging appears to be skin-specific and is defined as the effect of the external environment on the biological response of the skin. The skin's response to photoaging (which can be caused by exposure to the sun and pollution, as well as smoking) is often associated with a lack of normal hydration, the appearance of dilated capillaries (arach...
