Embodiment 1

[0053] The development and application of the microsatellite molecular marker of Castanopsis silk chestnut of the present invention go through the following steps:

[0054] 1. Obtaining the DNA sequence of Castanopsis similarius

[0055] Using the Latin name of Castanopsis fargesii (Castanopsis fargesii) as a key word, the gene sequence database maintained by the National Institutes of Health (GenBank: http: / / www.ncbi.nlm.nih.gov / ) was searched. The registered nucleotide sequence of Castanopsis chinensis is saved in the form of fasta file.

[0056] 2. DNA sequence processing of Castanopsis similarius and screening of DNA sequences containing microsatellites

[0057] The downloaded sequences were spliced by online splicing (CAP3) and redundant removal program (CD-HIT), and then the microsatellite analysis program (MISA) and the online microsatellite site analysis software (MicrosatelliteRepeats Finder) compiled in Perl language were used. The processed sequences were screen...

Abstract

The invention discloses specific primers and a detection method of Castanopsis fargesii and Castanopsis carlesii microsatellite markers. Eight pair of specific primers, of Castanopsis fargesii microsatellite molecular markers, suitable for Castanopsis carlesii are developed by utilizing an existing Castanopsis fargesii nucleotide sequence in a common database, applying a bioinformatics method and performing experimental verification, and base sequences are shown as SEQ ID NO.1-16. The invention further provides a detection method of EST-SSR molecular markers of Castanopsis fargesii and Castanopsis carlesii. The detection method of the EST-SSR molecular markers has the advantages of quickness, simplicity, stability and low cost, and a reference is provided for developing of EST-SSR molecular markers of other Castanopsis plants. By the specific primers and the microsatellite marker detection method, novel tools are provided for study on population genetic differentiation and structure, genetic diversity level of populations, gene flow among the populations and mating systems of the Castanopsis plants like Castanopsis fargesii and Castanopsis carlesii and study on molecular assisted selection of the Castanopsis plants.

Description

technical field [0001] The invention belongs to the technical field of plant genome DNA molecular markers, and in particular relates to specific primers and detection methods for microsatellite markers of Castanopsis sinensis and Castanopsis militaris. Background technique [0002] Microsatellite (microsatellite), also known as simple sequence repeat (SSR), refers to a DNA sequence repeated in series with 1 to 6 nucleotides as a unit, which is commonly found in eukaryotes and prokaryotes genomes and even viral genomes. SSR molecular markers have the following characteristics: repeat units and repeat numbers are highly variable, polymorphic information capacity is high; co-dominant, following Mendelian inheritance; flanking sequences are relatively conservative; selection neutral, etc. [0003] Based on the above characteristics, microsatellite molecular markers are widely used in many research fields such as species fingerprinting, genetic map construction, population genet...
