Abstract

The invention provides a method for identifying activity of mini-sized potato seeds with EESI-MS (extractive electrospray ionization-mass spectrometry) and a multivariate statistical technique. The method comprises steps as follows: to-be-tested mini-sized potato seed samples are subjected to artificial aging acceleration processing; the mini-sized potato seeds after artificial aging acceleration treatment are subjected to a germination standard test, and germination indexes and activity indexes are calculated; the mini-sized potato seeds subjected to aging acceleration treatment are selected through random sampling for preparation of extracting solutions, and supernatants are obtained; the supernatants of the mini-sized potato seed extracting solutions are subjected to EESI-MS analysis, and the mini-sized potato seeds with different activity indexes are identified in combination with a variable analysis method. According to the method, the mini-sized potato seed extracting solutions with different activity levels are subjected to mass spectrometric analysis, and the mini-sized potato seeds with different activity indexes are identified in combination with the variable analysis method, so that the method has the advantages of being good in stability, high in sensitivity, high in analysis speed and the like.
