Embodiment

[0057] The application of STAT1 promoter methylation detection in drug-resistant tumor cells includes the following steps:

[0058] (1) Establishment and identification of drug-resistant cells in vitro: construct drug-resistant glioma U251 cells and colon cancer SW480 cells in vitro, culture glioma cells and colon cancer cells in a specific medium for 6 months, and then use MTT method was used to detect drug-resistant glioma cells and colon cancer cells; the special medium for glioma U251 cells was purchased from Cyagen;

[0059] The method for constructing drug-resistant glioma U251 cells is as follows:

[0060] After the U251 cells were cultured for 1 week in a special medium containing a final concentration of 1 nM alisertib, the surviving U251 cells were resuspended in a special medium with a final concentration of 10 nM alisertib to continue culturing. The dedicated medium containing alisertib at a final concentration of 10 nM was replaced every 2 days. After 3 weeks, t...

Abstract

The invention relates to the field of biomedicine, provides an application of STAT1 promoter methylation detection in drug-resistant tumor cells, and solves the defect of no method for monitoring thedrug resistance to alisertib in the prior art. The application includes the following steps: (1) establishment and identification of in-vitro drug-resistant cells: constructing in-vitro drug-resistantbrain glioma cells and colon cancer cells, culturing the brain glioma cells and the colon cancer cells in a specific culture medium for 6 months, and detecting the drug-resistant brain glioma cells and colon cancer cells by an MTT method; (2) in vivo establishment of drug-resistant cells; (3) abnormal activation of STAT1 signaling pathways; and (4) promoter methylation detection.

Description

technical field [0001] The invention relates to the field of biomedicine, in particular to an application of STAT1 promoter methylation detection in drug-resistant tumor cells. Background technique [0002] Chemotherapy is one of the most important means of treating malignant tumors. However, the resistance of tumor cells to chemotherapeutic drugs often leads to the failure of chemotherapy and tumor recurrence. The new oral second-generation Aurora-A small molecule inhibitor alisertib (MLN8237) has the advantages of high targeting, low toxicity, and easy passage through the blood-brain barrier. Studies have found that it has a good effect on a variety of tumors. Currently, there are 59 clinical trials on alisertib worldwide; however, there is no method for monitoring resistance to alisertib. Contents of the invention [0003] Therefore, aiming at the above content, the present invention provides an application of STAT1 promoter methylation detection in drug-resistant tum...
