Abstract

The freehand sectioning method comprises the following steps of acquiringclean and fresh peony tuber tissues, dewatering the peony tuber tissues until the water content is 5-10 percent, cutting the obtained tuber tissues with a blade in a direction perpendicular to a longitudinal axis to acquire a section with the thickness being 20-30 microns, putting the section in absolute ethyl alcohol to remove excessive moisture, and putting the treated section in distilled water for complete unfolding. For the application of the freehand sectioning method, the section is put under a microscope for observation and photographing, and photographing is finished within 15 minutes after sectioning is finished. The freehand sectioning method provided by the invention is simple to operate, high in sectioning efficiency and low in cost, can thoroughly solve the problems of severe experiment condition, complex operation process, use of a toxic reagent, relatively long time and relatively large cost of experiment investment and the like of the existing paraffin sectioning method, is simple and available in used tools and low in cost, does not use the toxic reagent, is environmentally friendly, is wide in application and has an important theoretical and practical significance.
